Abstract
The influence of different nitrogen sources on the proliferation ofM. smegmatis is investigated in two deficient media (modified Kirchner substrate 30). The proliferation is measured by means of the oxygen uptake and the mass of cells; in certain cases the utilization of the substrates is estimated additionally.
Asparagine and glutamine are effective upon proliferation of the germs, the same effect is observed with glutamic acid but not with aspartic acid. Amides like urea, acetamide, nicotinamide and isonicotinamide increase the proliferation. The germs are also able to use nitrate and nitrite for their nitrogen metabolism in a limited degree.
The action of utilisable nitrogen sources on the proliferation is dependent on the concentration. In addition some more factors have an influence, e.g. the pH value of the medium in the presence of nitrite.
The combination of the substrates with limited amounts of asparagine is followed by an increase in the oxygen uptake and the mass of cells.
Zusammenfassung
Der Einfluß unterschiedlicher Stickstoffquellen auf die Vermehrungsvorgänge beiM. smegmatis wird in zwei Mangelmedien (modifiziertes Kirchner-Substrat 30 nach Herrmann) untersucht. Die Vermehrung wird über Sauerstoffaufnahme und Zellvolumen erfaßt; bei ausgewählten Ansätzen wird zusätzlich der Verbrauch der Substrate bestimmt.
Asparagin und Glutamin bewirken eine starke Vermehrung der Keime, von den entsprechenden Säuren zeigt nur Glutaminsäure cinen gleichen Effekt. Amide wie Harnstoff, Acetamid, Nicotinamid oder Isonicotinamid bewirken eine Förderung der Vermehrung; Nitrat und Nitrit können vonM. smegmatis in beschränktem Umfange zur Deckung des Stickstoffbedarfes herangezogen werden.
Die Wirkung der verwertbaren Stickstoffquellen ist konzentrationsabhängig. Darüber hinaus sind noch weitere Faktoren von Einfluß, wie beispielsweise der pH-Wert des Mediums bei Einsatz von Nitrit.
Werden die Substrate mit begrenzten Mengen von Asparagin kombiniert, so hat ihr Zusatz eine Erhöhung von Sauerstoffaufnahme und Zellvolumen zur Folge.
