Summary
Isoxyl tagged both with3H and35S is administered twice at the rate of 6 g to twelve volunteers; the blood levels are determined simultaneously on identical samples, by radiochemical and microbiological methods after administration of two pharmaceutical forms: Isoxyl “tablet form” and Isoxyl “powder form”. The two intakes of tagged product are separated by an interval of thirteen days in the course of which a normal cure of Isoxyl is established (6 g/day of cold product). The adopted protocol is of a cross-over type with supplementary controls. The results obtained by the two methods of determination show blood levels as high as 10 to 12 µg/ml which is practically twice as much as that observed in the animal (rabbit) treated in similar conditions.
The means obtained by the two methods of determination are in agreement except during the hour which follows the administration of the first half of the dose; indeed at this moment, radiochemistry detects inactive metabolites.
The comparison of the blood levels obtained after administration of Isoxyl “powder” form and Isoxyl “tablet” form shows in a significant way that the powder form improves or at least accelerates the resorption of the active product. This conclusion is reached with the radiochemical as well as with the microbiological determinations. Finally, the comparison of the blood levels determined on days 0 and 14 do not allow the detection of significant variations which could suggest an enzymatic induction or a phenomenon of accumulation.

De l'Isoxyl doublement marqué (3H et35S) est administré à raison de 6 g, à deux reprises à 12 volontaires; les taux sanguins sont déterminés simultanément, sur échantillons identiques, par les méthodes radiochimique et microbiologique, après administration de deux formes pharmaceutiques: Isoxyl „comprimés“ et Isoxyl „poudre“. Les deux prises de produit marqué sont séparées par un intervalle de 13 jours au cours duquel une cure normale d'Isoxyl est instaurée (6 g/jour de produit froid). Le protocole adopté est du type cross-over avec témoins supplémentaires. Les résultats obtenus par les deux méthodes de dosages permettent la mise en évidence de taux sanguins pouvant aller jusqu'à 10 à 12 µg/ml ce qui représente pratiquement le double de ce qui est observé chez l'animal (lapin) traité dans des conditions similaires. Les moyennes obtenues par les deux dosages sont concordantes sauf dans l'heure qui suit l'administration de la première moitié de la dose; à ce moment, en effet, la radiochimie détecte des métabolites inactifs.
La comparaison des taux sanguins obtenus après administration de l'Isoxyl „poudre“ et de l'Isoxyl „comprimés“ fait apparaître de manière significative que la forme „poudre“ améliore ou au moins accélère la résorption du produit actif. Cette conclusion est fournie aussi bien par les dosages radiochimiques que par les dosages microbiologiques. Enfin, la comparaison des taux sanguins déterminés aux jours 0 et 14 ne permet pas de détecter des variations significatives qui puissent faire penser à une induction enzymatique, ni à un phénomène d'accumulation.
Zusammenfassung
6 g doppelt markiertes (3H und35S) Isoxyl werden 12 Freiwilligen zweimal verabreicht; die Blutspiegel werden nach Verabreichung der beiden pharmazeutischen Formen „Isoxyl-Tabletten“ und „Isoxyl-Pulver“ an übereinstimmenden Proben simultan mit radiochemischer und mikrobiologischer Methode bestimmt. Die beiden Einnahmen von markiertem Produkt sind durch einen Zeitabstand von 13 Tagen getrennt, in deren Verlauf eine normale Isoxyl-Kur weitergeführt wird (6 g unmarkiertes Produkt pro Tag). Das gewählte Versuchs-Protokoll ist vom Typ cross over mit zusätzlichen Kontrollen. Die mit den beiden Bestimmungsmethoden erzielten Ergebnisse gestatten die Darstellung von Blutspiegeln, die von 10 bis 12 µg/ml gehen können, womit praktisch das Doppelte der beim Tier (Kaninchen) beobachteten Werte erreicht ist, das unter ähnlichen Bedingungen behandelt wurde. Die mit beiden Methoden erhaltenen Mittel stimmen überein, außer in der Stunde, die der Verabreichung der ersten Hälfte der Dosis folgt; zu diesem Zeitpunkt werden radiochemisch inaktive Metaboliten festgestellt.
Der Vergleich der nach Verabreichung von Isoxyl „Pulver“ und Isoxyl „Tabletten“ erhaltenen Blutspiegel zeigt in bemerkenswerter Weise, daß die Pulverform die Resorption des aktiven Produktes verbessert oder zumindest beschleunigt. Diese Konklusion ergibt sich sowohl aus den radiochemischen wie aus den mikrobiologischen Bestimmungen. Schließlich entdeckt der Vergleich der an den Tagen 0 und 14 bestimmten Blutspiegel keine signifikanten Abweichungen, die an eine enzymatische Induktion oder an ein Akkumulationsphänomen denken ließen.
