Summary
The influence of incubation-time, temperature and pH on the generation of the antigen-antibody-complex was investigated in the radioimmunoassay of angiotensin II. The optimal conditions are: incubation-time between 3 and 20 hrs at 4‡C and pH 7.4. Under these conditions it was found not only the highest degree of antigen-antibody-complex, but also the best reproducebility of the method. Higher temperature degrades the antibody bounded angiotensin by reversible inhibition or by enzymatic degradation. Dimercaprol or EDTA inhibits this enzymatic activity. Under optimal conditions no other substances need to be added to activate the reaction.
Zusammenfassung
Es werden die Einflüsse von Inkubationszeit, Temperatur und pH auf die Bildung des Antigen-Antikörper-Komplexes bei der radioimmunologischen Bestimmung von Angiotensin II untersucht. Als optimale Bedingung ergibt sieh eine Inkubationszeit zwischen 3 und 20 Std, eine Temperatur von 4‡C und ein pH-Bereich von 7,4. Unter diesen Bedingungen ist nicht nur die erreichbare Bindung von Angiotensin II am Antikörper am höchsten, sondern auch die Reproduzierbarkeit der Methode am sichersten. Die Erhöhung der Temperatur bringt eine Verringerung des vom Antikörper gebundenen Angiotensins teils durch reversible Hemmung der Komplexbildung, teils durch enzymatischen Abbau von Angiotensin, was durch Zusatz von enzymhemmenden Substanzen wie Dimercaptopropanol und EDTA verhindert werden kann. Besondere Zusätze zum Inkubationsansatz bei optimalen Bedingungen sind nicht erforderlich.
