Zur Prüfung der Cortisonwirkung auf die Kollagensynthese der Osteoblasten erhalten 30–40 g schwere weibliche Wistarratten insgesamt 70 mg Cortison in täglichen Dosen von 5 mg i. p. 30, 60 und 240 min vor dem Tode wird diesen Cortisonratten sowie den Kontrollratten eine einmalige Dosis von 250 μC3H-Prolin injiziert. Die proximale Tibiaepiphyse wird lichtmikroskopisch und autoradiographisch untersucht.

Die lichtmikroskopischen Veränderungen bestehen in einer Verschmälerung der Epiphysenfuge, einer verringerten Zahlder Osteoblasten, einer Irregularität der Trabekelbreite und in einer Verbreiterung der Zone der präparatorischen Verkalkung und der primären Spongiosa.

Die Osteoblasten weisen 240 min nach der3H-Prolininjektion noch eine deutliche Markierung auf, im Gegensatz zu denjenigen der Kontrollratten. Diese Veränderungen werden als eine Hemmung der Ausschleusung der Kollagenvorstufen gedeutet.

To test the effect of cortisone upon collagen synthesis of osteoblasts female Wistarrats weighing 30–40 g were given altogether 70 mg of cortisone in daily doses of 5 mg intraperitoneal. 30, 60 and 240 minutes before being killed these cortisonerats as well as the control rats are injected with a single dose of 250 uC3H-Proline. The proximal epiphysis of the tibia is examined lightmicroscopically and autoradiographically.

The lightmicroscopical changes are a diminution of the epiphyseal juncture, a diminished number of osteoblasts, an irregularity of the trabeculas, and an expansion of the zone of the calcification and the primary spongiosa. 240 minutes after the injection of3H-Proline the osteoblasts show a clear marking contrary to those of the control rats.

These changes are taken for a restraint of the extrusion of the tropocollagen.
