Zur Prüfung der Parathormonwirkung auf die enchondrale Ossifikation erhalten 40–60 g schwere Wistarratten in einer ersten Versuchsserie je 200 I E Parathormon i. p. Diese Ratten werden in Intervallen von 1 bis 48 Std nach Parathormoninjektion getötet, nachdem ihnen zusätzlich 40 min vor dem Tode eine einmalige Injektion von 25ΜC3H-Thymidin i.p. injiziert wurde.
In einer zweiten Versuchsserie erhalten alle Ratten zu Versuchsbeginn je 200 I E Parathormon und 50ΜC3H-Thymidin und werden in Intervallen von 1 bis zu 7 Tagen getötet.

Der3H-Index der intermitotischen Knorpelzellen liegt während der ganzen Versuchsdauer im Bereiche der Norm. Der3H-Index der Präosteoblasten steigt nach Parathormoninjektion von 7% innerhalb von 6 Std auf 14%, nach 12 Std auf 24% an und sinkt 48 Std nach Parathormoninjektion auf 10% ab. Die3H-Indices der Osteoblasten und Fibroblasten liegen während der ganzen Versuchsdauer zwischen 2 und 3%.

Eine mittlere Dosis von Parathormon führt zu einer 12–24 Std dauernden Proliferation der Präosteoblasten, aus denen sich in der nachfolgenden 4 bis 5tägigen Differenzierungsphase die anderen Knochenzellelemente ableiten. Diese Differenzierungsphase gliedert sich in eine initiale Knochenabbau- (12 Std nach Parathormoninjektion stärkste Osteoklasie) und in eine anschließende Aufbau-oder Knochenneubildungsphase (1. bis 4. Tag nach Parathormoninjektion).

Die Fibroblasten des Periostes bilden eine eigene Zellpopulation und weisen nach Parathormoninjektionen keine gesteigerte Teilungsaktivität auf.

To test the effect of parathormone upon endochondral ossification, a series of Wistarrats weighing 40–60 g each received 200 I.U. of parathormone i.p. These rats were sacrificed at intervals of 1 to 48 hours after administration of the parathormone and after an additional single injection of 25ΜC3H-thymidine i.p. was administered exactly 40 minutes prior to death.
A second series of rats received an initial dose of 200 I.U. parathormone each with a concurrent injection of 50ΜC3H-thymidine and were sacrificed at intervals of 1 to 7 days.

The3H-Index of the intermitotic cartilage cells lies within the normal range throughout the duration of the entire experiment. The3H-Index of the preosteoblasts rises within 6 hours after an injection of parathormone from a normal value of 7% to 14%, continues its rise to 24% after 12 hours and then decreases to 10% within 48 hours after the parathormone administration. The3H-Indices of the osteoblasts and fibroblasts remain during the entire experiment at values of 2–3%.

An intermediate dosage of parathormone leads to a preosteoblastic proliferation that lasts for 12 to 24 hours and from which evolve the other bone cells during the following 4–5 day differentiation phase. This differentiation phase may be subdivided into an initial phase of demolition (12 hours after parathormone administration osteoclasis is greatly increased) and a following phase of reconstruction or new bone formation (during the 1st–4th day after parathormone administration).

The periosteal fibroblasts form an individual cell population and exhibit no increase in mitotic activity after parathormone administration.
