Summary
Healthy mongrel dogs were anaesthetized with morphine-chloralose and loaded intravenously with glucose or glucose combined with tolbutamide. Bloodflow in the V. lienalis was measured and blood glucose concentrations were determined in plasma from V. lienalis and A. femoralis. From these data glucose output and uptake were calculated. There was evidence for glucose uptake in the normoglycemic as well as in the hyperglycemic state. Glucose output occurred only when the spleen was maximally constricted. It was concluded that the dog's spleen does not function as reservoir for glucose, but that the organ's need for glucose is great. In contrast to the authors, earlier results in the liver a mathematical relationship between arterial glucose level and glucose uptake was not demonstrable. In both groups (glucose load and glucose in combination with tolbutamide) bloodflow in the V. lienalis declined. This was explained in three ways: 1. loss of erythrocytes as a result of blood sampling, 2. excitation of volume receptors in the mesenteric circulation, 3. mobilisation of adrenaline in response to the insulin stimulating action of tolbutamide. Recording of bloodflow in the V. lienalis revealed typical rhythmic waves. With regard to their frequency and amplitude these waves were classified as the “resting” and “excitation” type.
