Summary
The immunosuppressive effect of cyclophosphamide and of histone extracted from calf thymus was compared using skin homotransplantation as experimental model in inbred as well as in randombred mice. The results obtained are the following:

Rejection of skin homograft is delayed by pretreatment of graft receptors with histone. A similar effect was observed when treatment was started at the fourth day after grafting. Treating the graft receptors with cyclophosphamide during the time between 1 day before and 2 days after transplantation rejection was also delayed. In these experiments the degree of delay was the same when using histone or cyclophosphamide. No additive effect was observed in combining the two kinds of treatment.

Intravenous pretreatment of graft receptors with only one injection of histone shortly before transplantation resulted also in a significant delay of skin homograft rejection. There was a linear dose-effect-ratio in the range of 0.78 to 100 μg histonenitrogen per dosis.

Significant delay in skin homograft rejection was also observed when graft donors were treated with histone. This was not the case using cyclophosphamide as immunosuppressive drug.

Injection of cyclophosphamide resulted in marked leucocytopenia. This was not observed even after repeated injections of histone. In this case the values of lymphocytes and polymorphonuclear cells remained normal.
Zusammenfassung
Der immunosuppressive Effekt von Cyclophosphamid und von Histon aus Kalbsthymus wurde am Modell der Haut-Homotransplantation bei Inzucht- und bei Nicht-Inzuchtmäusen verglichen. Die Versuche ergaben folgende Resultate:

Eine Vorbehandlung der Transplantatempfänger mit Histon bewirkt eine statistisch signifikante Verzögerung der Transplantatabstoßung. Ein ähnlicher Effekt wird beobachtet, wenn die Tiere erst ab dem 4. Tag nach der Haut-Homotransplantation mit Histon behandelt werden. Eine Behandlung mit Cyclophosphamid in der Zeit zwischen 1 Tag vor und 2 Tage nach der Transplantation verzögert ebenfalls die Transplantatabstoßung. Der Effekt von Histon und von Cyclophosphamid ist vergleichbar. Eine Kombination beider Behandlungsverfahren ergibt aber keine additive Wirkung.

Eine Hemmung der Transplantatabstoßung wird auch dann beobachtet, wenn den Transplantatempfängern Histon kurzfristig vor der Transplantation intravenös injiziert wird. Im Bereich von 0,78 bis 100 μg Histon-N besteht ein linearer Dosis-Wirkungs-Effekt.

Werden nur die Transplantatspender, nicht aber die Transplantatempfänger mit Histon vorbehandelt, dann haftet das Transplantat ebenfalls in statistisch signifikanter Weise länger. Dieser Effekt ist bei Verwendung von Cyclophosphamid nicht zu beobachten.

Wiederholte Injektionen von Histon führen im Gegensatz zu Cyclophosphamid nicht zu einer Leukocytopenie. Im Falle von Histon bleiben die Werte für Lymphocyten und Granulocyten im strömenden Blut im Normbereich, während es nach Cyclophosphamid zu einer ausgeprägten Lymphocytopenie und Granulocytopenie kommt.
