Summary
A cell-free system from rabbit reticulocytes incorporated radioactive leucine into the soluble protein. The incorporation depended on the maintenance of an optimal pH and required the presence of microsomes and a reagent mixture, containing an amino acid-mixture, ATP, GTP, GSH, and an energy-supplying system. Small quantities of RNase and puromycin inhibited the incorporation of leucine completely. The inhibition of the incorporation of leucine by several drugs depended on the concentration of the drugs. Experiments with phenylbutazone and chlorpromazine hinted at the possibility that these substances inhibited not only the incorporation of leucine into soluble protein but also its release from the microsome fraction.
Zusammenfassung
Ein zellfreies System aus Kaninchenreticulocyten baute radioaktives Leucin in das lösliche Protein ein. Der Leucineinbau war von der Einhaltung eines optimalen pH-Wertes abhängig und erforderte außer der Anwesenheit von Mikrosomen die Gegenwart einer Reagentienmischung, die eine Aminosäurenmischung, ATP, GTP, GSH und ein energielieferndes System enthielt. Durch geringe Mengen RNase und Puromycin wurde der Einbau vollständig gehemmt. Die Hemmung des Leucineinbaus durch Pharmaka ließ eine Abhängigkeit von der Konzentration erkennen. Versuche mit Phenylbutazon und Chlorpromazin ergaben Hinweise dafür, daß diese Substanzen außer dem Einbau des Leucins in das lösliche Protein auch die Ablösung der Radioaktivität von der Mikrosomenfraktion hemmten.
