Summary
Creatinuria during catabolism is not influenced by the function of the thyroid gland. It persists unchanged when the thyroid hormone production is stopped. Creatinuria is not due to a reduced consumption of creatine in the muscle but to an increased synthesis.
A reduced synthesis of protein is accompanied by an increase in creatine production. In stimulating the anabolism, as for instance by the androgen Methylandrostenolon, a decrease in the synthesis of creatine was observed.
It is demonstrated by these experiments that reducing or stimulating the protein synthesis leads to a contrary effect on the creatine synthesis. The creatine metabolism in the muscle remains unchanged under these conditions. The application of STH reduces the creatine synthesis by a stimulation of the protein synthesis, on the other hand it inhibits the creatine metabolism in the muscle. In the whole the production of creatinin is decreased.
Therefore, STH, as an anabolic substance causes as well creatinuria. The reason is its inhibitionary effect on the enzyme activity of creatine kinase.
Looking for the possibility to influence the creatine metabolism of the muscle by hormones we examined the efficacy of TSH. It was shown that TSH increases directly the synthesis of creatine without any influence on the protein metabolism. Therefore this effect has no relation to catabolism.
A normal creatinuria was observed in the athyroid animal inspite of the increase of creatine synthesis by TSH. This is caused by a greater consumption of creatine in the muscle. TSH raises the activity of creatine kinase and therefore augments the creatininuria.
Zusammenfassung
Bei Ausschaltung der Schilddrüsenhormonproduktion tritt die katabole Kreatinurie weiterhin unverändert auf. Ihre Entstehung ist auch nicht auf einen verminderten Kreatinverbrauch in der Muskulatur, sondern auf eine vermehrte Synthese zurückzuführen.
Ein verminderter Eiweißaufbau geht mit einer vermehrten Kreatinsynthese einher. Wird der Aufbauprozeß gesteigert, so geht die Kreatinsynthese zurück. Der durch den androgenen Stoff Methylandrostenolon hervorgerufene Anabolismus wird von einer herabgesetzten Kreatinsynthese begleitet.
Diese Untersuchungen zeigen daher, daß eine Hemmung oder Steigerung der Eiweißsynthese den umgekehrten Prozeß in der Kreatinsynthese mit sich bringt. Der Muskelstoffwechsel von Kreatin bleibt dabei unbeeinflußt. Es wird aber festgestellt, daß STH einerseits durch Steigerung des Eiweißaufbaues die Kreatinsynthese herabsetzt, andererseits hemmt es den Kreatinstoffwechsel in der Muskulatur, so daß die Kreatininproduktion abnimmt. Hierdurch wird nach Anwendung einer Substanz, die eine anabolische Stoffwechsellage bedingt, dennoch eine Kreatinurie hervorgerufen. Ihre Ursache ist auf eine durch STH bedingte Hemmung der Aktivität des Enzyms Kreatinphosphokinase zurückzuführen.
Auf der Suche nach einer hormonellen Beeinflussung des Kreatinstoffwechsels der Muskulatur wird die Wirkung des thyreotropen Hormons untersucht. Es stellt sich heraus, daß TSH direkt auf den Kreatinstoffwechsel einwirkt. Es steigert nämlich die Synthese des Kreatins, ohne in nennenswerter Weise den Eiweißstoffwechsel zu beeinflussen. Diese Wirkung erweist sich somit als unabhängig von dem Zustand des Aufbauvorgangs des Organismus und hat daher zum Katabolismus keine Beziehungen. Trotz der vermehrten Kreatinsynthese nach TSH wird beim athyreotischen Tier keine Verstärkung der physiologischen Kreatinurie festgestellt. Zur gleichen Zeit wird der Kreatinverbrauch der Muskulatur gesteigert; TSH wirkt nämlich als Enzyminduktor. Es steigert die Aktivität der Kreatinphosphokinase des Muskels, so daß Kreatinin vermehrt produziert wird.
