Zusammenfassung
Es wurden 16 Ratten untersucht, welche mit chronischen Elektroden im lateralen Hypothalamus eine hohe Frequenz der Selbstreizung aufwiesen. Der Reizstrom wurde alternierend «optimal» bzw. um 20% niedriger (= «suboptimal») dosiert. Die Selbstreizungsfrequenz bei optimalem Reizstrom (hM) betrug etwa 80–95/min; jene bei suboptimalem Reizstrom (nM) etwa 30–50/min. Die hM wurde weder durch Nikotin noch durch Amphetamin verändert. Die nM wurde durch 1 bzw. 2 mg/kg Amphetamin sowie durch 0,1, 0,25 und 0,5 mg/kg Nikotin in qualitativ gleicher Weise erhöht. Bei der höchsten Dosis Nikotin von 0,5 mg/kg ging der Erhöhung eine kurze Hemmung der Leistung voran. Der Nikotineffekt war von etwa der 5. bis zur 120. Minute nach der Applikation nachweisbar.
Summary
16 rats with chronically implanted hypothalamic electrodes yielding high rates of self-stimulation were used in this experiment. The amount of reward was changed by using alternatively an «optimal» current level and a 20% lower «suboptimal» current level. Self-stimulation frequency varied between 80–95 responses/min, at the optimal stimulation level (hM), and between 30–50 responses/min. at the suboptimal level (nM). Neither amphetamine nor nicotine changed the hM. The nM was increased by 1 respectively 2 mg/kg amphetamine and in a qualitatively very similar way by 0.1, 0.25 and 0.5 mg/kg nicotine. With the highest dose of 0.5 mg/kg nicotine the increase of performance was preceded by a short inhibitory phase. This nicotine effect could be noticed from about the 5th to the 120th minute after application depending on the dose level.
