Zusammenfassung
Es wird ein Überblick über Prinzipien zur Bestimmung der biologisch wirksamen Calciumkonzentration (ionisierter bzw. diffusibler Anteil) im Blutserum gegeben sowie eine einfache Methode zur kolorimetrischen Analyse des diffusiblen Calciums im Ultrafiltrat von 0,5 ml Serum (durch Cuprophanfolie) mit Chloranilsäure beschrieben (Standardabweichung ±3%). Die Erscheinung laufender Konzentrationsabnahme des diffusiblen Calciums mit fortschreitender Proteinkonzentrierung während der Ultrafiltration läßt sich, gestützt auf Modellversuche, als Folge einer vermehrten Calcium-Serumeiweiß-Bindung erklären. Es werden Serumnormalwerte des diffusiblen und Gesamtcalciums für verschiedene Species angegeben. Vergleichende Analysen der Calciumfraktionen im Serum gesunder, nebenschilddrüsenloser und calciumarm ernährter Ratten weisen auf einen weiteren Wirkungsmechanismus des Parathormons hin: der Bereitstellung von ionisiertem Calcium aus Calcium-Eiweiß-Verbindungen. Gesetzmäßige Beziehungen zwischen Gesamt-, diffusiblem Calcium- und Eiweißspiegel im Serum gesunder bzw. nebenschilddrüseninsuffizienter Ratten konnten nicht festgestellt werden.
Summary
Principles for estimation of biological active calcium concentration in blood and a simple method for colorimetrical analysis of diffusible calcium in serum ultrafiltrates are described. After filtering of 0.5 ml serum through “Cuprophanfolie” calcium is determined in ultrafiltrate with chloranilic acid (standard deviation ±3%). Model experiments interprete alterations of diffusible calcium concentration during filtration procedure in succession of increased calcium/serum proteine agglutination. Physiological values of diffusible and total serum calcium for a series of species are communicated. Compared analysis of this calcium fractionations in healthy, parathyreoidectomised and rats on low calcium diet indicate an additional mechanism of parathormone action. Parathyreoid glands seem to provide Ca2+ from calcium/proteine compounds. Lawful relations between total, diffusible calcium and protein content in serum can not made sure of healthy and parathyreoid insufficient rats.
