Bei Ratten führt eine mehrtägige intraperitoneale Zufuhr von Fe III-Polyfructose-Komplex zu einer geringgradigen Verminderung des Gesamteiweißgehaltes des Serums, zu einer hochgradigen Reduktion derγ-Globuline und einer Erhöhung derα-Globulinfraktion. In dieser Hinsicht ist eine relativ kurzfristige Behandlung mit hohen Dosen wirkungsvoller, als eine längerdauernde mit niedrigeren.

Histologisch läßt sich bei dieser Dysproteinämie eine hochgradige Eisenspeicherung der Zellen des RES in Leber und Milz nachweisen, die zur Zerstörung dieser Zellen führen kann. Die Schädigung der Parenchymzellen in der Leber ist weniger eindrucksvoll; in der Niere kommt es zum Auftreten einer Eiweißnephrose.

In der Humanpathologie finden sich ähnliche Serumeiweißkonstellationen bei der Antikörpermangelkrankheit und beim nephrotischen Syndrom. Da die Proteinausscheidung im Harn bei den mit dem Eisenkomplex behandelten Tieren geringfügig ist, wird angenommen, daß derγ-Globulinmangel durch eine Verminderung der Produktion dieser Fraktion zustandekommt. Offenbar ist dabei die Schädigung der Zellen des RES durch die excessive Eisenspeicherung von entscheidender Bedeutung.

The intraperitoneal administration through several days of a Fe III-polyfructose complex to rats leads to a small decrease of the total serum protein of the animals, a strong reduction of the gamma globulin level, and an increase of the alpha globulin level. Short-term treatment with high doses of Fe III-polyfructose is more effective than long-term treatment with lower doses.

Histologically the cells of the RE-system in liver and spleen are loaded with iron which may cause destruction of these cells. The damage to the parenchymal liver cells is much less impressive. The kidney shows signs of tubular nephrosis with protein casts in the tubules.

The serum protein pattern found in Fe III-polyfructose treated rats is similar to those in agammaglobulinemia and nephrotic syndrome in humans. Since there is no significant proteinuria in these animals, it may be postulated that the gamma globulin deficiency is the result of reduced synthesis. RES-cell damage by the excessive iron uptake seems to be of primary importance.
