Résumé
La distribution chez le lapin intact ou splénectomisé, d'une émulsion d'huile de paraffine (Paramul), injectée par voie intra-veineuse, et les lésions des filtres primaires du sang qui en découlent, sont étudiées. Ce n'est qu'après des quantités importantes de Paramul que les ganglions, filtres secondaires, sont atteints. Ce système réagit différemment à des substances exogènes, selon qu'elles sont administrées sous forme corpusculaire ou soluble.
Les images de début, avec leurs caractéristiques histologiques et histochimiques, permettent de démontrer la stimulation, puis l'épuisement du métabolisme des cellules ayant capté l'émulsion. Tout au cours de l'évolution, particulièrement après un intervalle prolongé après l'arrêt des injections, on peut suivre la redistribution des gouttelettes d'émulsion. Le foie est l'organe le plus gravement atteint, d'abord par la formation de foyers de captation, puis la cicatrisation de ceux-ci qui provoquent une fibrose de sa partie gauche, alors que la partie droite ne montre que des lésions peu prononcées.
Les faibles modifications que subissent les poumons sont attribuées à leur rôle d'organes évacuateurs. La moelle osseuse se signale par sa passivité à l'égard de l'émulsion, de même que la pulpe blanche de la rate, alors que la pulpe rouge participe à la formation de foyers de captation. Les ganglions lymphatiques ne manifestent que tardivement la captation de l'émulsion en produisant de petits foyers.
Aux stades ultimes de nos expériences on peut observer une paramyloïdose de la rate, des villosités intestinales et des reins.
Certains de ces résultats sont discutés en corrélation avec les dosages radiochimiques pratiqués à l'aide d'une émulsion marquée par le C14.
Zusammenfassung
Die Verteilung und der Einfluß auf die Blutfilter einer intravenös injizierten Paraffinölemulsion (Paramul) werden beim intakten und splenektomierten Kaninchen untersucht. Erst nach großen Dosen werden sekundär die Lymphdrüsen befallen. Die Blutfilter reagieren verschieden auf Substanzen, je nach ihrer corpusculären oder löslichen Form.
Die Anfangsstadien der Speicherung lassen histologisch und histochemisch eine Stimulation des Zellstoffwechsels erkennen. Die Spätbilder sprechen für eine allmählich einsetzende Zellerschöpfung. Eine Neuverteilung der Emulsionstropfen kann im Laufe der Experimente, besonders aber nach längerer Unterbrechung der Injektionen, beobachtet werden. Die Leber ist am stärksten betroffen, zuerst in der Form von Speicherungsherden, dann durch die nachfolgende Fibrose, die besonders die linke Hälfte befällt. Die Lungen sind wenig verändert, wahrscheinlich weil sie die Möglichkeit haben, die Emulsion schnell auszuscheiden. Das Knochenmark und die weiße Pulpa der Milz zeichnen sich durch ihre Passivität gegenüber der Emulsion aus; dagegen nimmt die rote Pulpa der Milz an der Bildung von Speicherungsherden teil.
In den Endstadien unserer Versuche können wir eine Paramyloidose der Milz, der Dünndarmzotten und der Nieren beobachten.
Einige Ergebnisse werden mit denen durch die mit C14 markierte Emulsion erhaltenen Resultate verglichen.
Summary
The distribution of an intravenously injected emulsion of paraffin oil (Paramul) given to intact or splenectomized rabbits, and the resulting effects on the primary filters of the blood, have been studied. The secondary filters, viz. the lymph glands, are concerned only after great quantities of Paramul. The reaction of this system against exogenous substances is different, according to their soluble or corpuscular state.
The initial lesions, with their histologic and histochemical characteristics, demonstrate the metabolic stimulation and the ensuing exhaustion of the cells having trapped the emulsion. The redistribution of the droplets can be seen all along the course of the experiments, especially after a prolonged interval of time following the interruption of the injections. The liver shows the most severe changes; first appear phagocytary nodules, the cicatrisation of which eventually produce the fibrosi of the left part of the liver; the right part is only moderately affected. The lungs, playing the part of evacuating organs, are only slightly involved. The bone marrow and the white pulp of the spleen remain remarkably passive to the emulsion; the red pulp on the contrary, takes part in the formation of phagocytary nodes. In the lymph nodes, phagocytary nodules appear very late.
Paramyloïdosis of the spleen, intestinal villi and kidneys may be seen in the latest stages of the experiment.
Some of these results are discussed in correlation with the radiochemical ones after injection of a14C-labeled emulsion.
