Zusammenfassung
Es wird eine einfache Mikromethode zur chemischen Serumproperdinbestimmung beschrieben. 0,1 ml Serum wird bei 16° mit Zymosan inkubiert, der Properdin/Zymosan-Komplex mit physiologischer NaCl-Lösung frei von reversibel adsorbierten Serumeiweißen gewaschen, bei 37° mit einem Veronalpuffer hoher Ionenstärke dissoziiert und das freigesetzte Properdineiweiß gegen ein Human-Testserum mit 6 E/ml und ein Human RP-Serum mit 0 E/ml nachLowry photometriert. Mit dieser Methode lassen sich die Properdintiter normaler, pathologischer und hämolytischer Seren von Mensch und Tieren mit einer Standardabweichung von ±8% bestimmen. Bei Ratten und Mäusen ist der Properdinspiegel auf diese Weise erstmalig am Einzeltier täglich meßbar. Für eine Reihe von Tierspecies werden physiologische Serumproperdinwerte mitgeteilt und mit serologisch nach einem modifizierten Verfahren vonMaassen gewonnenen Ergebnissen sowie Literaturwerten verglichen. Die chemische Mikromethode ist für umfangreiche Routineuntersuchungen unter Berücksichtigung verschiedener Kautelen geeignet.
Summary
A simple micro method is described for the chemical serum properdin estimation. 0,1 ml of serum is incubated at 16° with zymosan, the properdin/zymosan-complex with physiological NaCl-solution is washed free from reversibly adsorbed serum proteins, it is dissociated at 37° with a veronal buffer of high ionic strength and the set at liberty properdin protein is photometered against a human test serum with 6 E/ml and a human RP-serum with 0 E/ml according toLowry. With this method it is possible to determine the properdin titers of normal pathological and hemolytic serums of man and of animals with a standard deviation of ±8%. With rats and mice the properdin level is measurable in this way for the first time at the single animal. For a series of animal species physiological serum properdin values are communicated and are compared with results, gained serologically according to a modified method ofMaassen, and they are compared with literary values. The chemical micro method is suitable for extensive routine investigations taking into consideration a number of precautionary measures.
