Zellen aus Rinde und äußerer Markzone der Rattenniere wurden mit Hilfe des Enzyms Kollagenase isoliert.

Die Wirkung von Strophanthinzusatz zum Suspensionsmedium und der Effekt der Inkubation in Na-freier Lösung auf den Sauerstoffverbrauch der isolierten Zellen wurde im Warburg-Apparat untersucht.

Der Sauerstoffverbrauch isolierter Nierenrindenzellen in einer isotonischen Nährlösung mit einer Na-Konzentration von 140 mM/l betrug bei 37°C im Mittel 0,699 (σ=0,091) ml O2·g−1 · min−1 (Trockengewicht). Nach Zusatz von g-Strophanthin in einer Konzentration von 1 mg/ml sank der O2-Verbrauch um 45% auf im Mittel 0,374 (σ=0,039) ml O2·g−1 · min−1 (Trockengewicht).

In Na-freiem Suspensionsmedium war die O2-Aufnahme bei 25°C Badtemperatur gegenüber der Sauerstoffaufnahme in Na-haltigem Medium im Mittel um 44,8% herabgesetzt.

Strophanthinzusatz zu Na-freier Suspension bewirkte keine weitere Senkung der O2-Aufnahme.

Die Befunde deuten darauf hin, daß die Energiefreisetzung für den aktiven Na-Transport der isolierten Rattennierenzellen ausschließlich durch eine Na-aktivierte, strophanthinhemmbare ATPase gesteuert wird, und daß von den isolierten Zellen etwa 45% des aufgenommenen Sauerstoffs für den Na-Transport verbraucht werden.

The mean O2-uptake at 37°C of isolated cells in media containing 140 mM Na/l lay at 0.699 (σ=0.091) ml O2·g−1 · min−1 (dry wt.). Addition of Ouabain (1 mg/ml) induced a mean reduction of the O2-uptake of 45% to 0.374 (σ=0.039) ml O2·g−1 · min−1 (dry wt.).

Incubation in Na-free media reduced the mean O2-uptake by 44.8%.

Addition of Ouabain to Na-free media did not reduce the O2-consumption any further.

It is concluded that the liberation of energy for active Na-transport of isolated tubule cells of the rat is controlled by an ATPase which is activated by Na and inhibited by Ouabain, and that about 45% of the O2-uptake of the isolated cells in media containing Na in physiological concentration is used for the transport of sodium.
