Zusammenfassung
Es wird ein „Abgekürztes Verfahren” der Isolierung von Nematoden aus Pflanzenteilen beschrieben, mit welchem ein beträchtlicher Aufwand an Arbeitszeit erspart wird. Die Isolierung wird anstatt in reinem Wasser in 0,15%igem Wasserstoffsuperoxyd durchgeführt und dauert wie bei der bisher üblichen Methode zwei Tage. Man erreicht bei der Isolierung vonA. ritzemabosi aus Chrysanthemenblättern das 1,5–2fache und bei der Untersuchung von Erdbeerpflanzen aufAphelenchoides spp. dieselben Ergebnisse wie mit der bisherigen Methode. Eine Verlängerung der Extraktionszeit über die Dauer von zwei Tagen hinaus bringt bei der Isolierung in reinem Wasser keine weitere Steigerung der Ausbeute. Bei dem nicht gekürzten Verfahren in 0,15%igem H2O2 steigt die Isolierung in der zweiten oder dritten Woche der Extraktion nocheinmal an. Die Ausbeute kann hierdurch bei der Isolierung vonAphelenchoides spp. aus Erdbeerpflanzen bis auf das rund Achtfache gegenüber der bisherigen Methode oder dem „Abgekürzten Verfahren” ansteigen. — Der Einfluß der Probengröße auf die Untersuchungsergebnisse ist an einigen Beispielen aufgezeigt.
Summary
On the isolation of bud and leaf nematodes (Aphelenchoides fragariae andA. ritzemabosi) from strawberry and chrysanthemum. Part I
A simplified method for the isolation of nematodes from plants is described. 0,15% hydrogen peroxyde is used to replace tap water. Compared with the conventional methods, the amount ofA. ritzemabosi separated from chrysanthemum in 2 days is increased by 1,5 to 2 folds and the same amount, as in the usual methods, ofAphelenchoides spp. is obtained from strawberry plants. Using tap water, the amount of nematodes isolated does not increase by prolonging the extraction to more than 2 days. The nematodes are stacken due to oxygen deficiency. Using 0,15% H2O2, the amount of nematodes increases once more during the 2nd. or 3rd. week of the extraction (see Fig. 1). With this, the return can be increased up to 8 folds compared with the conventional methods, — The influence of the size of samples on the results is demonstrated with some examples.
