Zusammenfassung
Die magnetische Kernresonanzspektroskopie hat sich im Laufe der letzten Jahre zu einer attraktiven Methode für Studien der Molekülkonformationen von Proteinen in Lösung entwickelt. In mancher Hinsicht bildet sie eine ideale Ergänzung der Einkristallröntgen-untersuchungen. In der vorliegenden Arbeit wird versucht, die Beziehungen zwischen Proteinstruktur, Proteinkonformation und den verschiedenen NMR-Parametern anschaulich darzustellen. Anschliessend werden einige ausgewählte Themenkreise etwas eingehender diskutiert: Vergleich der Proteinkonformationen in Lösung und in Einkristallen, Denaturierung von Proteinen, dynamische Aspekte von Proteinkonformationen in Lösung, Studien von Enzym-Substratwechselwirkungen.
