Zusammenfassung
Zur Untersuchung der Zelleiweiß der Kurztag-Crassulacee Kalanchoë Bloßfeldiana wurde eine Methode entwickelt, die es gestattet, Blattproteine auch aus gerbstoffreichen Pflanzen zu isolieren.
Die Proteinfraktion der jüngsten Blätter wurde einer sauren Hydrolyse unterworfen; unter den Spaltprodukten konnten 24 ninhydrinpositive Substanzen aufgefunden werden; davon ließen sich 18 Komponenten mit bekannten Aminosäuren identifizieren.
Die aus der Spitzenregion derKalanchoë isolierten Blattproteine unterscheiden sich in ihrer Aminosäurezusammensetzung von den von anderen Autoren untersuchten Zelleiweißen aus den Dauergeweben höherer Pflanzen sowie von den bekannten Algenproteinen. Insbesondere ist der Anteil an basischen und cyclischen Aminosäuren gegenüber den Aminodicarbonsäuren deutlich erhöht. Diese Abweichungen sprechen für einen höheren Histongehalt — und damit vermutlich auch höheren Nucleoproteidgehalt — der untersuchten meristematischen Gewebe.
Während der photoperiodischen Induktion kommt es in der Spitzenzone zu einer Veränderung der Zelleiweiße die sich durch die quantitative Auftrennung der Hydrolyseprodukte erfassen läßt. Diese unmittelbar nach Induktionsbeginn eintretenden Verschiebungen im Mengenverhältnis der einzelnen Aminosäuren zeigen keine eindeutige Abhängigkeit von der Induktionsdauer; vielmehr durchlaufen die Konzentrationskurven innerhalb weniger Tage zwei mehr oder weniger deutliche Wendepunkte.
Die nach 30tägiger Induktion feststellbaren Eiweißveränderungen lassen sich nicht ohne weiteres auf die in den ersten Induktionstagen ablaufenden Prozesse zurückführen.
Die möglichen Zusammenhänge der beobachteten Eiweißumsetzungen mit Veränderungen im Enzymbestand der Zellen werden diskutiert.
