Zusammenfassung
Die Ergebnisse von Stoffwechseluntersuchungen isoliert mit Rinderblut perfundierter Rattenlebern werden mitgeteilt. Im Gleichgewicht stellt sich im Perfusionsmedium ein Lactat/Pyruvat-Quotient von etwa 10 ein. Unter diesen Bedingungen finden eine kontinuierliche Harnstoffsynthese und ein Abfall des anfänglich erhöhten Blutammoniakspiegels statt. Die Freisetzung von Glucose aus der Leber überwiegt deren Umsatz. Von der Durchblutungsgröße ist der L/P-Quotient abhängig, die Harnstoffsynthese jedoch nicht. Mit titriertem Lysin wurde die Synthese von Plasmaproteinen gemessen und der Umsatz von Lysin bestimmt.
Summary
Isolated livers from fed rats were perfused with a simple medium, consisting of bovine whole blood diluted with saline 5:1, heparine and oxytetracycline using Millers technique. The concentrations of glucose, lactate, pyruvate, ammonia and urea were estimated in the medium during 150 min.
First, glucose is released from glycogen stores suchlike lactate and pyruvate, probably due to preceeding anoxia. After 30–60 min the lactate/pyruvate ratio approximates 10, reflecting metabolic equilibrium. Then, only small changes in glucose concentration are observed. Reduced blood flow results in high lactate levels, increasing the L/P ratio. The initially high ammonia content is decreased and kept down by liver urea synthesis. Urea is produced continuously, independent from liver blood flow.
With tritiated 1-lysine and amino-acid addition, plasma protein synthesis, lysine turnover and oxydation can be estimated.
The results show, that in most instances, bovine whole blood is an appropriate medium for metabolic studies in rat-liver.
