Summary
Autoantibodies of Myasthenia gravis sera bind to an antigen belonging to the myosin complex of the sceletal muscle. These autoantibodies are organ specific but not species specific. Rabbits, immunized with the “myasthenic” antigen of human sceletal muscle fail to produce an autoantibody of the Myasthenia type, whereas rats do produce this autoantibody if immunized with rabbit “myasthenic” antigen.
There is a striking difference in the results of the Ouchterlony technique and indirect immunofluorescence technique, since immunofluorescence does not differentiate antibodies versus closely related antigens.
Production of myasthenic symptoms by immunization with muscle antigens did not occur in any of these animals. Microscopic slides of various organs of these animals did not exhibit any of the typical myasthenic alterations.
Zusammenfassung
Die Autoantikörper der Myasthenia gravis-Seren binden ein Antigen ab, welches zur Myosinfraktion des Skeletmuskels gehört. Diese Autoantikörper sind organspezifisch, aber nicht speciesspezifisch. Kaninchen bilden nach Immunisierung mit dem „Myasthenieantigen“ des menschlichen Skeletmuskels keinen Autoantikörper vom Myasthenietyp, während Ratten diesen Autoantikörper produzieren, wenn sie mit dem „Myasthenieantigen“ des Kaninchenskeletmuskels immunisiert werden.
Die Ergebnisse der Untersuchungen mit Hilfe der Agar-Gel-Doppeldiffusionstechnik und die Ergebnisse der Immunfluorescenz sind insofern unterschiedlich, als die Immunfluorescenztechnik es nicht erlaubt, zwischen nahe verwandten Antigenen bzw. deren Antikörpern zu unterscheiden.
Nach Immunisierung mit Muskelantigenen trat bei keinem der Tiere eine myasthenische Symptomatik auf. Mikroskopische Schnitte der verschiedensten Organe dieser Tiere zeigten keine Veränderungen, wie sie typisch für das Vorliegen einer Myasthenia gravis sind.
