Summary
The highest titers in antisera against homologous lymphocytes were found after the primary immunization with a skingraft and subsequent boostering with lymphocytes. C57Bl/6-anti-A sera had higher titers than A-anti-C57Bl/6 sera. Heterologous ALS and normal rabbit serum produced granulopenia. Homologous ALS decreased lymphocytes transiently, heterologous ALS permanently. Normal mouse serum, homologous ALS, even simple bleeding provoked granulocytosis.
Homologous ALS, especially when applied chronically, prolonged skingraft survival, though less than heterologous ALS. Pretreatment with homologous ALS potentiated the effect of heterologous ALS given after skingrafting. The combination of homologous ALS with normal rabbit serum increased the immunosuppressive effect of the former.
Zusammenfassung
Zur Herstellung hochtitriger homologer Antilymphocytenseren, deren Aktivität mit dem Dextranhämagglutinationstest gemessen wurden, erwies sich ein Verfahren am geeignetsten, das aus einer Grundimmunisation mit einem Hauttransplantat und Boosterungen mit Lymphocyten bestand. C57Bl/6-anti-A-Seren wiesen wesentlich höhere Hämagglutinationsspiegel auf als A-anti-C57Bl/6-Seren. Heterologes ALS, aber auch Kaninchennormalserum, verursachte deutliche Granulopenien. Homologe Antiseren führten zu vorübergehenden, heterologe Antiseren zu anhaltenden Lymphopenien. Alleinige Blutabnahme, Mäusenormalseren und homologe Antiseren führten zu Granulocytosen.
Homologes ALS zeigte, gemessen an der Überlebenszeit allogener Hauttransplantate, besonders bei intensiver Verabreichung eine deutliche immunsuppressive Wirkung, die aber schwächer war als nach Gabe von heterologem ALS. Eine Vorbehandlung mit homologem ALS verstärkte die immunsuppressive Wirksamkeit einer gleichgroßen Nachbehandlung mit heterologem ALS und erzielte den einer reinen heterologen Vor- und Nachbehandlung vergleichbaren Effekt. Homologes ALS hatte in Kombination mit heterologem Normalserum eine zusätzliche immunsuppressive Wirkung.
