Summary
The distribution of125I-iodinated rat albumin in the subcellular compartments of the rat liver (600 g, 3300 g, 12500 g, 100000 g and supernatant fractions) was studied 30 min (group I), 2 hrs (group II) and 4 hrs (group III) following intravenous injection. The PBI radioactivity is greatest in the supernatant, due mainly to the presence of blood. In the subcellular compartments, the concentration related to protein content in groups I and II, is greatest in 3300 g and 12500 g fractions. It was established that a considerable proportion of PBI radioactivity in the structure-bound fractions, present in the soluble form mainly in group I, can be precipitated with specific antibody.
Zusammenfassung
Die Verteilung des I125-markierten Rattenalbumins wurde in den subcellulären Kompartimenten der Rattenleber (600 g-, 3300 g-, 12500 g-, 100000 g- und Überstandsfraktion) 30 min (Gruppe I), 2 Std (Gruppe II) und 4 Std (Gruppe III) nach intravenöser Injektion untersucht. Die proteingebundene Radioaktivität ist im Überstand, vorwiegend durch Blutbeimengung, am höchsten. Unter den subcellulären Kompartimenten ist die Anreicherung bezogen auf Proteingehalt in den Gruppen I und II in den 3300 g- und 12500 g-Fraktionen am höchsten. Es wird festgestellt, daß sich ein nicht unerheblicher Anteil der PBI-Radioaktivität in den strukturgebundenen Fraktionen, die vorwiegend in der Gruppe I in löslicher Form vorhanden ist, mit spezifischem Antikörper präcipitiert werden kann.
