Summary
A perfusion system with continous dialysis of the perfusion medium was used in experiments with isolated rat livers. Metabolite content and bile secretion were determined as functional criteria.
Tissue content of ATP, ADP, AMP, lactate, pyruvate, glycerol-1-phosphate, dihydroxyacetonephosphate, malate, oxalacetate, fructose-1,6-diphosphate, glucose and glycogen as well as the ratios of lactate/pyruvate, glycerol-1-phosphate/ dihydroxyacetonephosphate and malate/oxalacetate correspond toin vivo conditions.
Dialysis of the perfusion medium results in a higher bile secretion rate of the isolated organ compared with common perfusion systems using a semisynthetic medium.
Advances and applications of the described perfusion system are discussed.
Zusammenfassung
Eine Perfusionsanordnung mit kontinuierlicher Dialyse des Perfusionsmediums wurde an der isolierten Rattenleber überprüft. Als Kriterien für den funktionellen Zustand wurden Metabolitgehalte und Gallesekretion herangezogen.
Die Gewebskonzentrationen von ATP, ADP, AMP, Lactat, Pyruvat, Glycerin-1-phosphat, Dihydroxyacetonphosphat, Malat, Oxalacetat, Fructose-1,6-diphosphat, Glucose und Glykogen sowie die Redox-Quotienten Lactat/Pyruvat, Glycerin-1-phosphat/Dihydroxyacetonphosphat und Malat/Oxalacetat entsprachen weitgehend Verhältnissen in vivo.
Die Dialyse des Perfusionsmediums bewirkt eine höhere Gallesekretion des isolierten Organs als bei Verwendung eines herkömmlichen Perfusionssystems mit halbsynthetischem Medium.
Vorteile und Anwendungsmöglichkeiten der beschriebenen Versuchsanordnung werden diskutiert.
