Zusammenfassung
Nach einer experimentell hervorgerufenen chronischen Osteomyelitis wird der Ablauf der Resorption des Amyloids durch neutrophile Leukozyten in Milz, Leber und Nieren bei der Maus (C 57 BL/6J) elektronenmikroskopisch untersucht. Die Resorption beginnt kurz nach dem Auftreten der ersten Amyloidablagerungen 8 Wochen nach der Operation. In den primären und tertiären Granula der neutrophilen Leukozyten wird als lysosomales Leitenzym die saure Phosphatase (EC 3.1.3.2) und in den spezifischen (sekundären) Granula die alkalische Phosphatase (EC 3.1.3.1) elektronenmikroskopisch bestimmt. Im Mittelpunkt der Untersuchungen steht die Frage nach dem Mechanismus der Kontaktaufnahme zwischen dem fibrillären Protein und den lytischen Enzymen. Die Aufnahme des Amyloids erfolgt in Membran-begrenzten Einschlüssen, die mit den Granula verschmelzen. Saure und alkalische Phosphatasen werden in diesen Phago-Lysosomen beobachtet. Nach dem Abschluß der Degranulation treten in einigen Einschlüssen Amyloidfilamente auf.
Summary
The stages of amyloid resorption by neutrophilic leukocytes following an experimentally induced chronical osteomyelitis are examined in spleen, liver and kidneys of mice (C 57 BL/6 J) electron microscopically. The resorption starts with the appearence of extracellular amyloid deposits eight weeks after operation. Acid phosphatase (EC 3.1.3.2) is determined as lysosomal marker-enzyme in the primary and tertiary granules and alkaline phosphatase (EC 3.1.3.1) in the specific (secondary) granules with the electron microscope. The main question is to find out how the fibrillar protein and the lytic enzymes come in contact. The engulfment of the amyloid occurs in membrane delimited inclusions which fuse with the granules. Acid and alkaline phosphatases are observed in these phago-lysosomes. After the process of degranulation amyloid filaments appear in some inclusions.
