Zusammenfassung
An 320 weiblichen Wistarratten wurde die Frage der potentiellen Nephrotoxizität von 7 Cephalosporinen untersucht. Die Antibiotika wurden 10 Ratten/Dosis und Substanz in 9 Einzeldosen an 5 aufeinanderfolgenden Tagen i. m. injiziert. Als Toxizitätsparameter dienten die Zellexkretionsraten für Nierenephitele sowie die Harnenzyme MDH, LDH und GOT, die vor, während und nach der Behandlung im 12-Stunden-Nachtharn bestimmt wurden. Zusätzlich erfolgten Serumharnstoffbestimmungen und histologische Untersuchungen der Nieren. Aus diesen Verlaufsanalysen waren unter Zugrundelegung des empfindlichsten Parameters, der Zellexkretion, für die einzelnen Substanzen folgende toxische Schwellendosen abzuleiten: Cephaloridin 500 mg/kg/Tag, Cephradin 1000 mg/kg/Tag, Cefazolin 2000 mg/kg/Tag, EMD 30087 2000 mg/kg/Tag, Cephacetril 3000 mg/kg/Tag, Cephalothin 3000 mg/kg/Tag, Cephapirin 5000 mg/kg/Tag. Der gewählte Untersuchungsgang gestattet somit, Cephalosporin-Antibiotika unter dem Aspekt Nierenverträglichkeit zu ordnen.
Summary
The question of the potential nephrotoxicity of 7 cephalosporins was investigated in 320 female Wistar rats. The antibiotics were administered to 10 rats per dose and substance in 9 separate i. m. doses on 5 consecutive days. The parameters for assessing toxicity were the rate of excretion of renal ephithelial cells, and the urinary enzymes MDH, LDH and GOT, which were determined before, during and after treatment in 12-hour urine voided overnight. In addition, serum urea was determined, and histological examination of the kidneys was performed. The following toxic threshold doses for the individual substances were derived from the analyses on the basis of cell excretion, the most sensitive parameter: Cephaloridine 500 mg/kg/day, cephradine 1000 mg/kg/day, cefazolin 2000 mg/kg/day, EMD 30087 2000 mg/kg/day, cephacetrile 3000 mg/kg/day, cephalothin 3000 mg/kg/day, cephapirin 5000 mg/kg/day. This type of investigation therefore permits cephalosporin antibiotics to be arranged in order of renal tolerability.
