Zusammenfassung
Zwei männliche Freiwillige erhielten einmalige intravenöse Dosen von 1,0 g beziehungsweise 0,9 g14 C-markiertem Cephacetril-Natrium. Beide Personen schieden innerhalb 24 Stunden 98 % der verabreichten Radioaktivität im Urin aus. Mittels inverser Isotopenverdünnungsanalyse der Urinproben konnte gezeigt werden, daß der Großteil der14 C-Dosis (im Durchschnitt 76%) als unverändertes Cephacetril eliminiert wird. Als wichtigster Biotransformationsschritt erweist sich die hydrolytische Spaltung der Esterbindung von Cephacetril, in dessen Folge Desacetyl-Cephacetril und/oder Lacton (21 %) im Urin auftritt. Die Spaltung der Amidbindung unter Bildung von Cyanessigsäure ist völlig unbedeutend (<1 %). Ein experimenteller Hinweis für das Entstehen von Cyanidionen liegt nicht vor.
Summary
Two male volunteers received single intravenous doses of14C-labelled cephacetrile sodium, 1.0 g being administered in one case and 0.9 g in the other. In both subjects, 98% of the administered radioactivity was eliminated in the urine within 24 hours. Inverse isotope dilution analysis of urine samples revealed that the greater part (on an average, 76%) of the14C-dose is excreted as intact cephacetrile. The principal biotransformation process consists in hydrolytic cleavage of the ester bond of cephacetrile, leading to the appearance in the urine of desacetyl-cephacetrile and/or lactone (21%). Hydrolysis of the amide bond involving the formation of cyanoacetic acid was insignificant (< 1%). There was no evidence to suggest that cyanide ions were formed.
