Zusammenfassung
In Versuchen an Mensch und Ratte, die zum Teil als Doppelblindversuche durchgeführt wurden, konnte mit statistischer Signifikanz folgendes für Dolantin gezeigt werden:

Es bewirkt dosisabhängig eine Hemmung der Wasserdiurese (Ratte, Mensch;p<1‰).

2.
Die absolute Urinionenexkretion in mval ist ebenfalls dosisabhängig vermindert (Ratte, Mensch;p<1‰).

3.
Die Urinionenkonzentration (mval/l) kann gesteigert, gleichbleibend als auch vermindert sein (Mensch;p<1‰).

4.
Der Effekt kommt nicht über eine Kreislaufwirksamkeit (verminderter peripherer Widerstand mit Blutdrucksenkung) zustande (Ratte).

5.
Der Dolantineffekt ähnelt dem Vasopressineffekt a) in Eintrittslatenz und Verlauf (Mensch und Ratte) und b) in der — allerdings nur bei einem Teil der Frauen beobachteten — Urinionenkonzentrations-steigerung.

6.
Der Effekt kommt nicht durch eine Erschlaffung der glatten Muskulatur des harnableitenden Systems mit Restharnbildung (Pscudoantidiurese) zustande (Mensch).

7.
Die Wirkung von 50 mg Dolantin entspricht der von 30 mE Vasopressin.
Von den fünf in der Literatur diskutierten Wirkungsme-chanismen des Dolantin konnte der vasculäre (Blutdrucksenkung) und der muskuläre (Erschlaffung des harnableitenden Systems) ausgeschlossen werden, es verbleiben — abgesehen vom suprarenalen Angriffspunkt (Aldosteronmechanismus) — noch der renale (direkter Angriff am Glomerulum oder Tubulus) und der hypophysär-renale Angriffspunkt (Vasopressin-mechanismus). Diese Frage soll durch einen biologischen ADH - Nachweis aus peripherem Blut in weiteren Versuchen geklärt werden.
Diese Ergebnisse sollten vor Dolantingaben in Situationen wie septischem Schock, Eklampsie, Niereninsuffizienz und postoperativ-anurischen Schmerzzuständen, in denen die Aufrechterhaltung bzw. das Wiederingangkommen der Diurese von lebenswichtiger Bedeutung ist, berücksichtigt werden.
Summary
Of the five mechanisms of action of Meperidine discussed in the literature (v. s.); the vascular (hypotensive action) and the muscular one (relaxation of the urinary tract) can now be excluded. Two mechanisms — apart from the adrenocortical one (aldosterone-mechanism) — still may exist: the renal one (direct effect either, on the glomeruli or on the tubules, or both) and the pituitary-renal one (vasopressin-mechanism). We will investigate these problems in further experiments by a biological estimation of antidiuretic hormone in peripheral venous blood after concentrating the posterior pituitary hormone by means of Sephadex-chromatography.
The experiments, which partially were performed as double blind tests, showed the following effects of Meperidine (Pethidine, Dolantin) in women and in rats:

it causes a dose-dependent water-antidiuresis (both in women and in rats;p<0,1%);

(2)
the absolute excretion of urinary ions (mEq.) is diminished in a dose-dependent manner (both in women and in rats;p<0,1%);
-
(3)
the concentration of urinary ions (mEq./l) can be increased, unchanged or diminished;
-
(4)
it has no circulatory effect;
-
(5)
the Meperidine effect is similar to that of vasopressin (a) in latency and course, and (b) in the increase in the concentration of urinary ions;
-
(6)
the effect of Meperidine in women is not caused by a relaxation of the smooth musculature of the urinary tract, with subsequent retention of urine (pseudo-antiduresis); and
-
(7)
the effect of 50 mg of Meperidine corresponds to that of 30 milliunits of vasopressin.
The administration of Meperidine (Pethidine, Dolantin) in situations such as septic shock, eclampsia, renal insufficiency and surgical pains with anuria, in all of which conditions the output, and maintenance thereof, of diuresis are of vital importance, should only be done under special care and precautions.
