Zusammenfassung

Der Watson-Schwartz-Test auf PBG fällt bei Tryptophanausscheidungen im Urin über 5 mg-% positiv aus. Tryptophanaldehyd verhält sich beim Ausschütteln mit Chloroform und Butanol wie PBG-aldehyd und zeigt ein Extinktionsmaximum bei ∼542,5 mµ (PBG-aldehyd bei ∼555 mµ). Tryptophan vermag daher einen PBG-Nachweis vorzutäuschen oder zu cachieren.

Summary

Watson-Schwartz-test for prophobilinogen (PBG) becomes false positive in tryptophanuria as seen in tryptophan loading tests.

Tryptophan is an Ehrlich reactor and its aldehyde is a red compound, chloroform insoluble like PBG aldehyde and also indistinguishible from PBG aldehyde by means of Butanol and Sodium acetate.

The spectrometric E max of tryptophan aldehyde is ∼542,5 mµ, of PBG aldehyde ∼555 mµ. In thin layer chromotography the RF of tryptophan was ∼0,67, of PBG ∼0,6.
