Zusammenfassung
Das geschwindigkeitsbestimmende Enzym bei der Inaktivierung von Angiotensin II durch menschliches Plasma ist wahrscheinlich eine Aminopeptidase. Wir beschreiben eine Methode zur Bestimmung der Aktivität dieses Enzyms. Menschliches Serum wird mit einem Überschußα-L-Angiotensin II-Amid inkubiert. Das dabei gebildeteα-L-Asparagin wird mit einer Asparaginase aus Meerschweinchen-Plasma inα-L-Asparaginsäure umgesetzt. Die Asparaginsäure wird mit einem DPN·H-abhängigen Photometer-Test bestimmt.α-L-Angiotensin II wird langsamer als das Amid gespalten. Wahrscheinlich werden die beiden Octapeptide durch verschiedene Aminopeptidasen abgebaut.β-L-Angiotensin II wird durch Aminopeptidasen des menschlichen Plasmas nicht gespalten.
Summary
The rate-limiting enzyme for the inactivation of angiotensin II by human plasma is probably an aminopeptidase. A method for the determination of the activity of this enzyme is described. Human serum is incubated with an excess ofα-L-angiotensin II-amide. Theα-L-asparagin formed by the aminopeptidase is converted to aspartic acid by asparaginase from guinea-pig plasma. Aspartic acid is measured by application of an DPN·H-dependent photometer-test.α-L-angiotensin II is split slowlier than the corresponding amide. Probably, the two octapeptides are inactivated by different aminopeptidases.β-L-angiotensin II is not split by aminopeptidases of human plasma.
