Zusammenfassung
498 mitPl. berghei infizierte Swiss-Mäuse wurden am 13. Inf. T. in sieben Gruppen geteilt und unterschiedlich behandelt. Die höchste Anzahl überlebender Mäuse wurde bei der Kombination von PABS-freier Nahrung (Milchdiät) mit oral verabreichtem Pyrimethamin (1–mg/kg) erzielt (58,6%). 43% überlebende Tiere wurden bei ausschließlicher Ernährung mit Milch und 36,1% mit oraler Pyrimethamintherapie bei gleichzeitiger Ernährung mit Standardfutter beobachtet.
Durch Zufuhr von p-Aminobenzoesäure (PABS) im Trinkwasser wird nicht nur die therapeutische Wirkung der allein verabreichten PABS-freien Diät vollkommen aufgehoben, auch die mit Pyrimethamin behandelten Tiere sterben dann ausnahmslos, ohne daß der Infektionsverlauf von den unbehandelten Kontrollen abweicht.
Es wird angeregt, bei Infektionen mit sulfonamidempfindlichen Erregern die exogene PABS-Zufuhr einzuschränken, um vielleicht hierdurch den chemotherapeutischen Index zu verbessern.
Summary
498 Swiss-mice infected with Pl. berghei were divided into 7 groups on the 13th day of infection and treated differently. The highest percentage (58,6%) of surviving mice was obtained by feeding p-aminobenzoic acid (PABA) free diet (milk diet) and additional pyrimethamine (1–2 mg/kg). 43% of the animals survived if they were fed exclusively with a milk diet and 36,1% by feeding with a standard food diet and oral pyrimethamine therapy.
Addition of PABA to the drinkwater leads to a complete suppression of the therapeutical effect of the milk diet and of the pyrimethamine treatment. In these cases the course of the infection does not differ from the controls.
It is suggested to lower the exogen PABA intake during infections with sulfonamid sensitive germs in order to improve the chemotherapeutical index.
