Summary

The influence of diazoxide on glucagon-stimulated insulin secretion has been studied using adrenalectomized rats. 200 mg/kg of diazoxide were injected intraperitoneally 2 hours prior to the experiments, and the animals killed by decapitation 1 or 5 minutes after intravenous injection of 0.1 mg/kg of glucagon. In animals fed ad libitum diazoxide considerably decreased insulin secretion stimulated by glucagon. After fasting for 24 hours, however, diazoxide showed no effect whatsoever on glucagon-stimulated insulin secretion. It has been concluded that the mode of action of glucagon on insulin secretion is different from that of glucose that has been shown to be blocked by diazoxide.
