Zusammenfassung

Die Ergebnisse der Leukocytenzählung mit Hilfe des Coulter Counters sind nicht nur von dem Zeitpunkt nach dem Saponinzusatz, an dem die Zählung vorgenommen wird, und der Schwelleneinstellung, sondern in erheblichem Maße auch von der Temperatur, bei der gearbeitet wird, abhängig. Vor allem bei Temperaturen über 30° sinken die Leukocytenzahlen stark ab, so daß Leukocytosen nicht erkannt und Leukopenien vorgetäuscht werden. Es ist erforderlich, das Gerät bei konstanter Temperatur zu eichen und alle späteren Zählungen bei dieser Temperatur durchzuführen, um verläßliche und reproduzierbare Leukocytenwerte zu erhalten.

The results of leucocyte counts by way of the Coulter Counter are not only dependent upon the period of time after the addition of saponin at which the counting is done also to a considerable degree upon the temperature at the working site. Especially at temperatures of 30° Celsius and over the figures of leucocytes are decreasing heavily, so that leucocytosis cannot be found any more and leucopenias are simulated. It is necessary to gauge the instrument on a constant temperature and have all later counts done at this particular temperature in order to obtain precise and reproducible figures of leucocytes.
