Zusammenfassung
An sechs stoffwechselgesunden Personen wurden während einer intravenösen Glucoseinfusion (1 ml einer 50%igen Lösung pro Minute, Infusionsdauer 190 min) die Umsatzrate der freien Fettsäuren (FFS) des Plasmas und die Einbaurate der Plasma-FFS in Plasmatriglyceride untersucht. Weiter wurde bei denselben Personen an einem der folgenden Tage der Einfluß einer intravenösen Infusion von Noradrenalin (0,25/min/kg) auf die genannten Stoffwechselgrößen während der Glucosebelastung untersucht, wobei mit der Verabreichung von Noradrenalin 60 min nach dem Start der Glucoseinfusion begonnen wurde. Der FFS-Umsatz und der Einbau der Plasma-FFS in Plasmatriglyceride wurden durch konstante Infusion von Palmitinsäure-9,10-H3 bestimmt, zur Berechnung der Umsatzgrößen wurde der Zeitraum von 130–190 min nach Beginn der Glucoseinfusion bzw. 70–130 min nach Beginn der Infusion von Noradrenalin herangezogen. Die Ergebnisse wurden mit den entsprechenden Werten einer Kontrollgruppe von elf Personen ohne medikamentöse Beeinflussung sowie den Werten von 16 Normalpersonen während einer Noradrenalininfusion in derselben Dosierung, jedoch ohne gleichzeitige Glucosebelastung, verglichen.
Die Glucosebelastung bewirkte eine signifikante Abnahme des FFS-Spiegels im arteriellen Plasma sowie eine signifikante Verminderung der Umsatzrate der FFS im Plasma und der Einbaurate der FFS des Plasmas in Plasmatriglyceride; die „Fractional turnover rate“ wurde durch die Verabreichung von Glucose nicht beeinflußt.
Während die Verabreichung von Noradrenalin allein eine signifikante Erhöhung des FFS-Spiegels im arteriellen Plasma und der Umsatzrate der FFS im Plasma sowie eine geringe, jedoch nicht signifikante Erhöhung der Einbaurate der FFS in Plasmatriglyceride bewirkte, wurde dieser Noradrenalineffekt durch die gleichzeitige Verabreichung von Glucose völlig aufgehoben: Zwischen der Abnahme des FFS-Spiegels, der FFS-Umsatzrate und der Einbaurate der Plasma-FFS in Plasmatriglyceride unter der Wirkung der Glucoseinfusion und den entsprechenden Werten während der Infusion von Glucose und Noradrenalin ließ sich kein Unterschied nachweisen. Auch die „Fractional turnover rate“ wurde durch die zusätzliche Verabreichung von Noradrenalin nicht verändert.
Für stoffwechselgesunde Personen ohne medikamentöse Beeinflussung sowie während der Glucosebelastung mit oder ohne zusätzliche Verabreichung von Noradrenalin ließ sich eine positive Korrelation zwischen der FFS-Konzentration im arteriellen Plasma und der Umsatzrate der FFS im Plasma nachweisen.
Summary
1) The turnover rate of plasma free fatty acids and the esterification rate of plasma free fatty acids to plasma triglycerides were estimated in six normal individuals by means of constant infusion of palmitic acid-9,10-H3 during an intravenous infusion of glucose (0.5 g/min) or glucose and norepinephrine (0.25 µg/min/kg).
2) Glucose loading results in a significant decrease of plasma free fatty acid level, a significant decrease of the turnover rate of plasma free fatty acids, and a significant decrease of the esterification rate of plasma free fatty acids to plasma triglycerides. The fractional turnover rate of plasma free fatty acids was not altered by the administration of glucose.
3) The effect of norepinephrine on the metabolism of free fatty acids in plasma (increase of concentration, increase of turnover rate, and a slight increase of the esterification rate) was completely abolished by the glucose load: The date of the individuals receiving glucose plus norepinephrine did not differ significantly from those obtained during glucose infusion alone.
4) The correlation between plasma free fatty acid concentration and turnover rate found in normal persons applied as well to individuals receiving a glucose load.
