Zusammenfassung
Isolierte menschliche Blutplättchen wurden auf Proteinase-Aktivität im neutralen und sauren Bereich untersucht. Ferner wurden sieben Exopeptidase-Aktivitäten bestimmt. Proteinase-Aktivität ließ sich nur im sauren Bereich (pH 3,5) nachweisen, aber mit verschiedenen Methoden eindeutig sichern. Mit synthetischen Substraten wurde die Anwesenheit von Kathepsin A wahrscheinlich gemacht. Die Exopeptidasen wurden hinsichtlich pH-Optimum (Aminotripeptidase), Aktivierbarkeit durchl-Cystein (katheptische Carboxypeptidase) und Metallionen (übrige Exopeptidasen) charakterisiert. Ihr Verhalten stimmte im wesentlichen mit den homologen Enzymen der Erythrocyten und Leukocyten überein. Die Aktivitäten der Proteinasen und katheptischen Enzyme lagen um 1–2 Größenordnungen niedriger als die der im Bereich von pH 7–8 wirksamen Exopeptidasen. Ein Vergleich von Thrombocyten, Erythrocyten, Leukocyten, Lymphocyten und Serum auf Volumenbasis zeigt, daß die Blutplättchen im allgemeinen hohe proteolytische Aktivitäten aufweisen. Auffällig ist besonders ihr Reichtum an katheptischen Enzymen, der für die Biologie und Pathophysiologie der Thrombocyten bedeutsam sein dürfte.
Summary
Proteolytic enzymes in normal human blood platelets. Isolated blood platelets were investigated for proteinase activity at neutral and acid pH. Furthermore, seven exopeptidase activities were estimated. Proteinase activity could only be demonstrated in the acid range (pH 3.5), but there it was confirmed by several different methods. By means of synthetic substrates, the presence of cathepsin A was made probable. The exopeptidases were characterized with respect to optimal pH (aminotripeptidase), activation byl-cysteine (catheptic carboxypeptidase), and metal ions (other exopeptidases). Their properties resembled essentially those of the homologous enzymes in erythrocytes and leucocytes. The proteinases and catheptic enzymes were by 1–2 orders of magnitude less active than the exopeptidase that are maximally active at pH 7–8. The enzyme activities of platelets, erythrocytes, leucocytes, lymphocytes, and serum are compared on a volume basis. The platelets in general show the highest activities. This is especially true for the catheptic enzymes which, therefore, may play an important role in the biology and function of blood platelets.
