Zusammenfassung
Angeregt durch eine Arbeit vonGerm undKubelka (1964) wird das Plasma der Rindenparenchymzellen des Stämmchens vonSelaginella martensii phasenoptisch untersucht. Der Organellenbestand entspricht dem einer normalen Pflanzenzelle. Da anscheinend jegliche Plasmaströmung fehlt, dürften die langen fädigen Mitochondrien oft eine Eigenbewegung beobachten lassen. Die vonGerm undKubelka als „Plasmanetze“ beschriebenen Konfigurationen erweisen sich als Plasmaleisten, welche durch die Vergrößerung von ER-Elementen in die Vakuole vorgepreßt werden.
Nach Zentrifugierung ist das Plasma von Spirogyrazellen einer Phasenkontrastbeobachtung besser zugänglich. Neben den meist kokkenförmigen Mitochondrien findet man, besonders zahlreich in älteren Zellen, leukoplastenähnliche Körper, deren Funktion noch ungeklärt ist. Genaue Analysen der Plasmaströmung lassen den Schluß zu, daß bei schwacher Strömung das dreidimensionale Netz des ER für die Strömungsbahn richtunggebend oder aber auch als submikroskopische Barriere für die Bewegung wirken kann. Starke Strömungsschübe zerteilen das Netz, und die ER-Elemente werden in ihnen parallelisiert. Die intraplasmatischen Vakuolen, die in der beschriebenen Ausbildung phasenoptisch sichtbare Teile des ER darstellen, zeigen in der Plasmaströmung das gleiche Verhalten.
Die schlauch- und vesikelförmigen intraplasmatischen Vakuolen bilden in der Spirogyrazelle ein sehr dynamisches Muster. Ihre Entstehung wird durch die verschiedensten Reize (Druck, Licht, Hitze, Plasmolytika), besonders auch durchα-Bestrahlung, induziert. Die Ausbildung ist, wie in einer Bildserie festgehalten wurde, reversibel, wobei lange dünne „Schläuche“ und Lakunen oft auch spontan im Plasma zu finden sind. Bläschen entstehen dagegen meist durch Zerfall und Abkugelung dieser „Schlauchelemente“ nach Alteration der Zellen. Bei Auftreten der intraplasmatischen Vakuolen scheint eine änderung der Permeabilitätseigenschaften der ER-Membranen zu erfolgen. So wird eine Hydratation und somit eine Volumsvergrößerung der in der Regel massearmen IZP (intrazisternale Phase) möglich, wodurch phasenlichte „Löcher“ im Plasma auftreten. Die änderung der Permeabilitätseigenschaften der ER-Membranen könnte durch eine Störung der Eiweißauflage des bimolekularen Lipoidfilms hervorgerufen werden. Bei der Vergrößerung des ER zu den intraplasmatischen Vakuolen wird dann nicht nur die Membran gedehnt, sondern es werden wahrscheinlich auch neue Moleküle eingebaut und so die Membranfläche vergrößert. Diese Vermutung scheinen auch Versuche mitα-Strahlen zu bestätigen.
Summary
A phase optical investigation into the plasma of cortex parenchyma cells ofSelaginella martensii stems was suggested by a paper ofGerm andKubelka (1964). The organelle content is in accordance with that of a normal plant cell. Movement of the long, threadlike mitochondria seems to be active, as there is an apparent lack of protoplasmic streaming. Configurations described as “protoplasmic nets” byGerm andKubelka prove to be protoplasmic ridges extruded into the vacuole by an enlargement of ER elements.
The protoplasm ofSpirogyra cells is more accessible to phase contrast observations after centrifugation. Besides mitochondria, chiefly of coccoid form, leucoplast-like bodies of yet unknown function are found, most numerously in older cells. From exact analyses of protoplasmic streaming we may conclude that in the case of slow streaming the threedimensional network of the ER may direct the course of the flow or may even form a submicroscopic barrier for it. Intensively streaming protoplasm breaks up the network, forcing ER elements to become parallel. Intraplasmic vacuoles, being phase optically visible parts of the ER in the form described, react in the same way to protoplasmic flow.
Tubular and vesicular intraplasmic vacuoles form a highly dynamic pattern inSpirogyra cells. Their formation is induced by most different stimuli (pressure, light, heat, plasmolyzing agents and, in particular, alsoα-rays). Their development is reversible, as documented by a series of photomicrographs: long, thin “tubes” and lacunae are often formed spontaneously in the plasma. Vesicles, on the other hand, develop by disintegration and rounding of these “tubular elements” after alteration of cells. Appearance of intraplasmic vacuoles seems to be accompanied by a permeability change in ER membranes. This allows hydration and volume increase of the ICP (intracysternal phase), which is of low mass as a rule. “Holes” of high transmittency for phase light are thus formed. Permeability changes of ER membranes may be a consequence of disorders in the protein coating of the bimolecular lipid film. During the enlargement of the ER to the final form of intraplasmic vacuoles the membranes not only extend, but their surface is probably also enlarged by the incorporation of new molecules. Experiments withα-rays also seem to confirm this conjecture.
