Summary
The new aminoglycoside antibiotic, sisomicin, was used to treat 19 hospitalized patients with a variety of infections caused by gram-negative bacilli. Sisomicin was administered for 9 to 44 days in a dose of 3 mg/kg/24 hours by the intramuscular route in divided doses every 8 hours. In all eight patients with urinary tract infection the organism was eradicated by therapy. A more variable response was seen in patients with osteomyelitis and soft tissue infection, attributed in part to underlying disease that was often encountered. The pharmacology of sisomicin was found to be similar to that reported for gentamicin. Adverse effects possibly attributable to the drug were noted in four patients, but were mild and reversible.
Zusammenfassung
Das neue Aminoglykosid-Antibiotikum Sisomicin wurde bei 19 Krankenhauspatienten mit verschiedenen Infektionen durch gramnegative Keime eingesetzt. Sisomicin wurde 9 bis 44 Tage in einer Tagesdosis von 3 mg/kg KG intramuskulär, verteilt auf drei Einzeldosen, gegeben. Alle acht Patienten mit Harnwegsinfektionen wurden durch die Behandlung keimfrei. Bei Osteomyelitis und Weichteilinfektionen war der Erfolg, abhängig von der Grundkrankheit, unterschiedlich. Die Pharmakologie des Sisomicin ähnelt der des Gentamicin. Nebenwirkungen, die möglicherweise dem Präparat zuzuschreiben sind, fanden sich bei vier Patienten; sie waren leicht und bildeten sich zurück.
