Summary
The interaction of antibiotics,Staphylococcus aureus and human polymorphonuclear leucocytes has been examined. Bacteria which had been exposed to benzylpenicillin, but not killed, became more susceptible to the bactericidal effects of leucocytes in vitro. Bacteria phagocytized by leucocytes from normal individuals and patients with chronic granulomatous disease were protected from the antibacterial effect of benzylpenicillin and gentamicin. Large numbers of phagocytized bacteria remained viable for 16–20 hours exposure to antibiotic concentrations that killed 98% or more non-phagocytized bacteria in four hours. In contrast, rifampicin caused a marked reduction of viable intracellular bacteria, indicating that this antibiotic penetrates better into the phagocytic vacuole than benzylpenicillin and gentamicin.
Zusammenfassung
Es wurde die Wechselwirkung zwischen Antibiotika,Staphylococcus aureus und menschlichen polymorphkernigen Leukozyten untersucht. Bakterien, die einem Kontakt mit Benzylpenicillin ausgesetzt gewesen, aber nicht abgetötet waren, wurden in vitro empfindlicher gegenüber den bakteriziden Wirkungen von Leukozyten. Durch Leukozyten von Gesunden und Patienten mit chronischen Granulomatosen phagozytierte Bakterien wurden gegen den antibakteriellen Effekt von Benzylpenicillin und Gentamicin geschützt. Große Mengen phagozytierter Bakterien bleiben bei einer 16–20stündigen Exposition gegenüber Antibiotika-Konzentrationen lebensfähig, die 98% oder mehr nichtphagozytierter Bakterien binnen vier Stunden abtöteten. Dagegen bewirkte Rifampicin eine erhebliche Reduktion lebensfähiger interzellulärer Bakterien; dieses Antibiotikum dringt daher anscheinend besser in die Phagozytenvakuole ein als Benzylpenicillin und Gentamicin.
