The ultrastructure of premelanosomes and melanosomes in eumelanin-producing cells and keratinocytes

Zusammenfassung

Die Bildung von Melanosomen in Eumelanozyten und ihre Einlagerung in Keratinozyten ist an der Embryonaldune der Wachtel (Coturnix coturnix) licht- und elektronenmikroskopisch untersucht worden.

Aus den Praemelanosomen mit einer Matrix aus mehreren konzentrisch geordneten und in sich gefalteten Membranen werden Melanosomen mit gleichförmiger Struktur. Sie bestehen aus einer trilaminaren Hüllmembran, die durch einen optisch leeren Spalt vom glatt konturierten Melanin getrennt wird. Das Melanin weist nach der Osmiumtetroxydfixation einen elektronendichten Saum mit feingranulärer Struktur auf, während das Zentrum mit und ohne Nachfixation homogen und weniger dicht erscheint.

Diese Feinstruktur bleibt erhalten, wenn die Melanosomen durch Ausläufer zu den Epidermiszellen transportiert werden. Die daran sich anschließende Verteilung auf das Zytoplasma ist regellos; sie endet mit der diffusen Einlagerung von Keratinfibrillen.

Summary

The formation of melasomes in eumelanocytes and their deposition in keratinocytes has been examined in the down feather of quail (Coturnix coturnix) embryos by light and electron microscopy.

Melanosomes of uniform structure are formed from the premelanosomes which have a matrix of multiple concentrically-arranged and folded membranes. The melanosomes are composed of a trilaminar membrane which is separated from the smoothly-contoured melanin by an optically empty gap. Following osmium tetroxide fixation, the melanin shows an electron-dense border with a fine granular structure whereas the centre appears homogeneous and less dense, both with and without such fixation.

The fine structure is maintained when the processes transport the melanosomes to the epidermal cells. The consequent distribution in the cytoplasm is irregular and terminates in a diffuse deposition of keratin fibrils.
