Methodological implications of testing anaerobe susceptibility to cephalosporins (cefazolin, cefamandole, cefoxitin)

Methodologische Implikationen der Empfindlichkeitstestung von Anaerobiern gegen Cephalosporine (Cefazolin, Cefamandol, Cefoxitin)

Summary

By simultaneously performing broth dilution, agar dilution, and agar diffusion tests withBacteroidaceae andPeptococcaceae, the influence of methodology upon the outcome of susceptibility testing to cefazolin, cefamandole and cefoxitin was studied. With beta-lactamase positive and negativeBacteroidaceae, the results of the broth dilution and agar dilution tests were in good agreement for cefoxitin. However, when tested with cefazolin and cefamandole, beta-lactamase positiveBacteroides strains had mostly high broth dilution MICs and relatively low agar dilution MICs. The statistical analysis of the relationship between zone size and broth dilution or agar dilution MICs frequently showed lack of stochastic linearity or relatively low correlation coefficients.

Zusammenfassung

Durch simultane Durchführung von Bouillon- und Agardilutions-sowie Agardiffusionstests mitBacteroidaceae undPeptococcaceae wurde der Einfluß der Methodologie auf das Ergebnis der Empfindlichkeitsprüfung gegenüber Cefazolin, Cefamandol und Cefoxitin untersucht. Bei Beta-Laktamase-positiven und -negativenBacteroidaceae bestand zwischen den Ergebnissen der Bouillon-und Agardilutionsversuche mit Cefoxitin gute Übereinstimmung. In Austestung gegen Cefazolin und Cefamandol zeigten Beta-Laktamase-positiveBacteroides-Stämme jedoch meist hohe Bouillondilutions- und relativ niedrige Agardilutions-MHK-Werte. Die statistische Bewertung der Beziehung zwischen Hemmhofdurchmesser und Bouillon- bzw. Agardilutions-MHK wies häufig fehlende Linearität oder relativ niedrige Korrelationskoeffizienten nach.
