Zusammenfassung
Es wird über die proteolytischen Veränderungen des Albumins durch das Fermentgemisch Bromelin berichtet. In einem bestimmten Konzentrationsbereich spaltet das Bromelin das Albumin in 2 Fraktionen, die sich voneinander durch ihre elektrophoretische Mobilität und hinsichtlich ihres Antigendeter minantengehaltes unterscheiden. Während die Fraktion I die gleiche elektrophoretische Wanderungsgeschwindigkeit wie das Originalalbumin aufweist, wandert die Fraktion II elektrophoretisch langsamer. Eine kurzzeitige Bromelineinwirkung führt zu 2 Albuminfraktionen, die immunchemisch nicht identisch sind. Durch eine mindestens 24stündige Inkubation mit dem proteolytischen Enzymgemisch entstehen 2 Albuminfraktionen, die eine immunchemische Verwandtschaftsreaktion zeigen. Bei einer Antigenverwandtschaft mit dem Originalalbumin hat die Fraktion I und noch mehr die Fraktion II durch die Enzymbehandlung Antigendeterminanten eingebüßt. Die Fraktion II besitzt weniger Antigendeterminanten als die Fraktion I.
Summary
The author reports on the proteolytic changes of albumin which are produced by the enzyme mixture Bromelin. In a certain concentration range Bromelin splits albumin into 2 fractions which differ from one another by their electrophoretic mobility and by their contents of antigen determining factor. Whereas fraction I has the same electrophoretic migration rate as the original albumin, the electrophoretic migration rate of fraction II is slower. A brief period of contact with Bromelin results in 2 albumin fractions which immunochemically are not identical. Incubation with the proteolytic enzyme mixture for at least 24 hours produces 2 albumin fractions which demonstrate a related immunochemical reaction. If the antigen is related to the original albumin then fraction I and even more, fraction II has lost antigen determining factors by the enzyme treatment. Fraction II has fewer antigen determining factors than fraction I.
