Zusammenfassung
Der photometrische Plättchenaggregationstest (PAT) wird beeinflußt von der Thrombozytenzahl und der Aktivität plättchenaggregierender Plasmafaktoren. Eine Thrombozytenzahl im Plasma von 150 000/mm3 sollte bei Anwendung dieses Testes nicht unterschritten werden.
Der photometrische PAT ist besonders geeignet zur Erfassung der spontan gesteigerten Aggregation im Plasma von Gefäßkranken. Die Wirkung aggregations-hemmender Stoffe wie Bencyclan (10−5molar) der Pyrimido-Pyrimidin-Verbindungen VK 744 (2×10−5molar) und SH 869 (5×10−6molar) sowie Acetylsalicylsäure (ASS) (2×10−4molar) konnte mit diesem Test gut erfaßt werden. In vivo ließ sich unter dem Einfluß 200 mg VK 744 i.v. eine kurzfristige und nach oraler Gabe von 1 g ASS eine 2 bis 6 Tage anhaltende aggregationshemmende Wirkung nachweisen.
150 γ SP54/ml führen nur in Gegenwart von Plasmaproteinen zu einer Aktivierung der Plättchenaggregation.
Summary
The photometric PAT is influenced by the platelet count and the activity of platelet-aggregating plasma factors. For reliable results the platelet count should not be less than 150 000 mm3. The test is sensitive, well reproducible and especially suitable for the estimation of spontaneously enhanced aggregation in the plasma of patients with vascular diseases.
Bencyclan (s. p. 252) (10−5molar), the pyrimido-pyrimidine compounds VK 744 (s. p. 252) (2×10−5molar) and SH 869 (s. p. 252) (5×10−6molar) as well as ASA (s. p. 252) (2×10−4molar) showed an inhibitory effect on platelet aggregation in the new test system.
In vivo a short lasting inhibitory effect on platelet aggregation was found after 200 mg of VK 744 i.v. Following the ingestion of 1 g ASA the inhibitory effect lasted for 2 to 6 days.
After addition of 150 γ SP54/ml (s. p. 252) to platelet rich plasma platelet aggregation is enhanced.
Plasmaproteins are essential for this effect.
