Zusammenfassung
Im Anschluß an ein Verfahren, das die gleichzeitige Isolierung von C\(\bar 1\)-Inaktivator und Inter-α-Trypsininhibitor erlaubt, gelang es uns, nun auch das RBP in reiner Form darzustellen und erstmals zu kristallisieren.
In 3 Schritten, die auf 3 verschiedenen Fraktionierungsprinzipien beruhen, nämlich: Ionenaustausch-Chromatographie, Löslichkeit in Ammoniumsulfat und Gelfiltration, wird zunächst der RBP-PA-Komplex isoliert. Durch Dissoziation des Komplexes und nachfolgende Gelfiltration in 6M neutraler Harnstofflösung wird reines RBP erhalten. Dieses kristallisiert mit Hilfe von Mineralsalzen wie Ammoniumsulfat, Natriumsulfat oder Kaliumphosphat in einem pH-Bereich von 5–8 in Form von langen Nadeln. Damit eröffnet sich die Möglichkeit der Strukturaufklärung mit Hilfe der Röntgenanalyse.
Die physikalisch-chemischen und chemischen Analysen des kristallisierten RBP stehen im Einklang mit den aus der Literatur bekannten Daten.
Summary
RBP has been isolated in a pure form and for the first time crystallized by using a method which permitted the simultaneous isolation of C\(\bar 1\)-Inactivator and Inter-Alpha-Trypsininhibitor.
First of all the RBP-PA complex was isolated after 3 steps based on different fractionation principles, viz.:- Ion-exchange Chromatography, Solubility in ammonium sulphate and Gel-filtration. The pure RBP was obtained after dissociation of the complex in 6M neutral urea solution and further gelfiltration. It crystallized out as long needles with help of mineral salts such as ammonium sulphate, sodium sulphate and potassium phosphate in a pH range from 5–8. The structural elucidation using X-Ray analysis is now possible.
The physico-chemical and chemical analyses of the crystallized RBP are in accord with the data already published.
