Scanning electron microscopy of erythrocyte ghosts prepared with and without ATP addition

Summary

The effect of ATP addition on the shape of erythrocyte ghosts prepared from fresh and stored human blood was studied by means of scanning electron microscopy. The following shapes of ghosts could be distinguished:biconcave ghosts resembling original biconvave erythrocytes, but with much flatter centres;smooth ghosts with no apparent biconcavity;crenated ghosts, the crenation ranging from slight to very heavy.

It was possible to prepare mainly biconcave ghosts from fresh erythrocytes sometimes even in the absence of ATP; sometimes they appeared exclusively after the addition of ATP. Stored erythrocytes without ATP gave rise mainly to crenated ghosts and with ATP mostly to biconcave ones. If subjected to repeated stress (for instance repeated changes in the tonicity of sucrose incubation medium), the ghosts from fresh blood had mostly crenated shape — favourable effect of ATP on their shape being only a negligible one.

Zusammenfassung

Mit Hilfe des Rasterelektronenmikroskops wurde der Einfluß des Zusatzes von ATP auf die Form der Erythrozytenschatten, die aus frischem und aus konserviertem menschlichem Blut gewonnen worden waren, untersucht. Die Schatten wiesen folgende Grundformen auf:Bikonkave Schatten, die den ursprünglichen Erythrozyten glichen, deren Mitte jedoch flacher war;glatte Schatten, die nicht als bikonkav erkennbar waren; schwach bis stark geschädigtegekerbte Schatten, auf denen Ausläufer von verschiedener Länge ersichtlich waren.

Aus frisch entnommenen Erythrozyten war es möglich, auch in Abwesenheit von ATP überwiegend bikonkave Schatten herzustellen, ein anderes Mal entstanden bikonkave Schatten nur nach dem Zusatz von ATP. Aus den konservierten Erythrozyten bildeten sich in Abwesenheit von ATP vorwiegend gekerbte Schatten, in Anwesenheit von ATP vorwiegend bikonkave Schatten. Wenn die Schatten aus frisch entnommenem Blut einer wiederholten Belastung unterworfen worden waren, z. B. einer wiederholten Variierung der Tonizität von Saccharose-Inkubationsmedium, entstanden überwiegend geschädigte gekerbte Schatten und ein Zusatz von ATP zu diesen Schatten übte nur einen ganz geringen günstigen Einfluß auf deren Form aus.

In der vorliegenden Arbeit wird über die Bedeutung der ATP und über die mögliche Bedeutung einiger ATPasen bei der Erhaltung der bikonkaven Form der menschlichen Erythrozyten diskutiert.
