Zusammenfassung
Die Untersuchung von Erythrozyten, die zur Desintegration der Membranen und zur Isolierung von Rh-Antigenen mit Ultraschall (12 kHz, 5 min) behandelt und “homogenisiert” worden waren, hat zu folgenden Ergebnissen geführt. Die RhAntigene (C und E) sind offenbar an größere funktionelle Bezirke der Erythrozytenmembran gebunden. Nach Schallbehandlung werden durch das Lysat mehr homologe, spezifische Antikörper neutralisiert als durch die gleiche Menge intakter Erythrozyten, was durch die Freilegung von am intakten Erythrozyten nicht zugänglicher Determinanten erklärt werden kann. Durch Immunisierung von Kaninchen mit dem nativen Schallhomogenat lassen sich keine Rh-spezifischen Antikörper stimulieren.
Summary
In order to desintegrate the membranes and to isolate Rh antigens erythrocytes with well defined Rh patterns are treated with ultrasonic waves. The results of investigation of the homogenous sonicates allow the following tentative conclusions: The Rh antigens (C and E) are combined obviously with certain functional membrane areas of a given size. After sonication the activity of the sonicate to neutralize specific antibodies is greater than the activity of the same amount of untreated cells. It is supposed that this effect is caused by making accessible hidden antigens that are inaccessible in the intact membrane. Immunization of rabbits with the sonicates does not stimulate Rh-specific antibodies.
