Zusammenfassung
Es wird über autoradiographische, radiométrische und elektronenoptische Untersuchungen an mit Phytohämagglutinin in vitro stimulierten Lymphozyten aus dem peripheren Venenblut klinisch gesunder Personen berichtet. Die Untersuchungen zeigten zunächst, daß die stimulierten Zellen in vitro eine radioaktiv markierte Aminosäure mit zunehmender Kultivierungsdauer in immer größerem Umfang aufnehmen, während nicht stimulierte Zellen nur eine geringe Markierung erfahren. Es fand sich weiterhin, daß der überwiegende Teil der Radioaktivität bei elektrophoretischer Auftrennung von Zell-Lysaten im kathodischen Bereich gefunden wird.
An Hand elektronenoptischer Aufnahmen von Kulturzellen wird gezeigt, daß die transformierten Lymphozyten keine Ähnlichkeit mit Plasmazellen besitzen, andererseits mit zunehmender Transformation in immer größerem Umfange Polyribosomen ausbilden, woraus geschlossen werden kann, daß die Zellen eine erhebliche Proteinsyntheseleistung vollbringen.
Summary
The author reports autoradiographic, radiometric and electronmicroscopic tests on lymphocytes from the peripheral venous blood from clinically healthy persons which had been stimulated in vitro with phytohemagglutinin. Initially the tests showed that the stimulated cells in vitro have a continuously increasing uptake of a radioactively labelled amino acid with increasing culture time. Cells that were not stimulated on the other hand only showed slight uptake of the labelled amino acid. Furthermore, the tests showed that the major part of the radioactivity is found in the region of the cathode on electrophoretic separation of cell-lysates.
On the basis of electronmicroscopic pictures of culture cells it is shown that the transformed lymphocytes do not have any similarity to plasma cells. On the other hand these transformed lymphocytes show a continuous increase of polyribosome formation with increasing transformation and from this one may conclude that the cells perform considerable protein synthesis.
