A simple method for measuring red cell deformability in models of the microcirculation

Summary

By measuring the flow rate of 10% red blood cell suspensions and of clear plasma through Nuclepore® sieves, the deformability of human red blood cells can be quantified. The relative flow rate (\(\dot V_{rel} \)) of red cell suspensions through the 5 μm pores at a driving pressure of 50 mm H2O is a direct measure of the red cell deformability in geometrical and dynamical models of the living microcirculation.\(\dot V_{rel} \) is about 0.7, it is largely independent of temperature between 4°C and 37°C, but highly dependent upon the pH of the sample. Reduction of\(\dot V_{rel} \) can be found in blood from patients suffering from various diseased states.

Zusammenfassung

Durch Messung der Fließrate einer 10%igen Suspension von roten Blutzellen (RBZ) und von klarem Plasma durch Nuclepore®-Siebe kann die Deformierbarkeit der menschlichen RBZ quantitativ bestimmt werden. Die relative Fließrate der RBZ-Suspensionen durch die 5 μm-Poren bei einem Filtrationsdruck von 50 mm Wassersäule ist ein direktes Maß der RBZ-Deformierbarkeit im geometrischen und dynamischen Modell der lebenden Mikrozirkulation.\(\dot V_{rel} \) beträgt ca. 0,7 und ist weitgehend unabhängig von Temperaturen zwischen 4°C und 37°C, dagegen hochgradig vom pH-Wert der Probe.

Reduziertes\(\dot V_{rel} \) kann im Blut von Patienten mit verschiedensten Krankheitszuständen beobachtet werden.
