Zusammenfassung
Bei Kaninchen wurde in verschiedenen Zeitabständen nach parenteraler Gabe von Kobaltchlorid und Natrium-Kobalt-III-Glukonat der Kobaltgehalt des Plasmas, der Erythropoetin-Titer des Plasmas, die59Fe-Inkorporation in die Erythrozyten, der Serumeisenspiegel und die totale Eisenbindungskapazität untersucht.
Der Kobaltspiegel des Plasmas lag nach Gabe beider Verbindungen so niedrig, daß eine Beeinträchtigung der Erythropoetintestung des Kaninchenplasmas an hungernden Ratten ausgeschlossen war. Kobalt wurde nach Gabe von Kobaltchlorid in dem bobachteten Zeitraum nach einer einfachen Exponentialfunktion aus dem Kreislauf eliminiert.
16 Stunden nach der Kobaltinjektion wurde bei kleinen Dosen, 12 Stunden danach bei hohen Dosen eine maximale Vermehrung des Erythropoetingehaltes im Plasma festgestellt.
Die Eiseninkorporation in die Erythrozyten zeigte 36–48 Stunden nach der Kobaltapplikation die höchste Einbaurate.
Aus der engen zeitlichen Verknüpfung dieser beiden Effekte wird gefolgert, daß die direkte Wirkung von Kobalt im Knochenmark auf einer Steigerung der Empfindlichkeit von Knochenmarkzellen gegen Erythropoetin beruht.
Der Serumeisenspiegel wird 8–12 Stunden nach Zufuhr von Kobalt wahrscheinlich durch einen Abstrom von Eisen in die Depotorgane erheblich reduziert. In den nächsten Stunden kommt es zu einem Wiederanstieg mit nachfolgendem erneuten weniger ausgeprägten Abfall innerhalb der erfaßten 48 Stunden. Die totale Eisenbindungskapazität ändert sich nicht.
Bei den beobachteten Wirkungen von Kobalt auf die Erythropoese konnten wir keinen Unterschied zwischen gleichen Dosen Kobaltchlorid und Natrium-Kobalt-III-Glukonat feststellen.
Summary
The changes in plasma content of cobalt and erythropoietin, incorporation of radioactive iron into erythrocytes, serum iron concentration and total iron binding capacity were studied in rabbits at various intervals after injection of cobalt chloride and sodium-cobalt-III-gluconate.
The erythropoietic effect of rabbit plasma in starved rats was not attributable to residual cobalt since the amount of cobalt remaining in the plasma after administration of the two substances was too small. Cobalt has been eliminated from the circulation in the observed interval after the injection of cobalt chloride according to a plain exponential function.
16 hours after administration of cobalt at lower dose levels and 12 hours afterwards at highers doses the peak of erythropoietin in plasma was reached.
The direct effect of cobalt on the incorporation of iron into red cells attained its maximum at 36 to 48 hours after the injection.
According to the chronological order of both actions the direct effect of cobalt in bone marrow has been explained as enhancing the responsiveness of bone marrow cells towards erythropoietin.
Serum iron concentration was reduced considerably 8 to 12 hours following cobalt administration probably due to flow into storage organs. In the next few hours serum iron concentration increased again and decreased not as significantly during the rest of the observed 48 hours. There has been no change of total iron binding capacity.
Identical doses of cobalt chloride and sodium-cobalt-III-gluconate caused comparable effects on erythropoiesis.
