Zusammenfassung
Es werden Befunde über die Gerinnungsaktivität von Hämolysat in vitro mit Hilfe des Rekalzifizierungs-Thrombelastogramms dargestellt. Die Reaktionszeit r und die Gerinnselbildungszeit k im Thrombelastogramm mit frischem Thrombozyten-armem Kaninchenplasma werden durch Hämolysat aus gewaschenen menschlichen und Kaninchen-Erythrozyten das 1–20 Tage bei −20° gelagert hat, signifikant stärker verkürzt als durch Hämolysat, das 30–90 Tage bei −20° gelagert hat. Die Irrtumswahrscheinlichkeit liegt unter 0,1%.
Es finden sich keine signifikanten Unterschiede zwischen der Wirkung von Hämolysat aus menschlichen und aus Kaninchen-Erythrozyten auf die Werte im Thrombelastogramm mit frischem und gefrorenem Thrombozyten-armem Kaninchenplasma.
Bei etwa 80% der Versuche steigt die maximale Thrombuselastizität me nach Hämolysatzusatz zu frischem Thrombozyten-armem Plasma und zu Plasma, das zuvor 24–48 Stunden bei −20° gelagert hat im Vergleich zu Nacl-Kontrollwerten an. Die Streuung der Einzelwerte ist erheblich, so daß sich der Effekt statistisch nicht sichern läßt.
Nach Zusatz von Hämolysat zu Thrombozyten-reichem Kaninchenplasma kommt es zu wenig ausgeprägten Veränderungen im Thrombelastogramm.
Summary
The results of the coagulation activity of haemolysate in vitro are demonstrated with the aid of the recalcification thrombelastogram. The reaction time r and the coagulation time c in the thrombelastogram carried out with fresh rabbit plasma of low thrombocyte content, are significantly reduced if a haemolysate of washed human and rabbit erythrocytes kept at −20° for 1–20 days is used, if compared with a haemolysate that had been kept 30–90 days at a temperature of −20°. The probability of error can be assessed as under 0.1%. There are no significant differences between the effect of an haemolysate from human and one from rabbit erythrocytes on the values arrived at in a thrombelastogram with fresh or frozen rabbit plasma of low thrombocite content.
In about 80% of the experiments the maximal thrombus elasticity me after haemolysate has been added to the plasma with low thrombocyte content and plasma that has been stored at −20° for 24–48 hours increases when compared with NaCl control values. The scatter of individual readings is considerable so that the effect cannot be statistically secured.
On adding the haemolysate to rabbit plasma which has a high thrombocyte content, no significant changes were observed in the thrombelastogram.
