Summary
The enhancing effect of two kinds of treatments combined with ALS on chronic secondary disease was studied. The strongly H2-incompatible donorrecipient combination C57BL/6J to CBA/J was used because antilymphocytic serum (ALS) in this combination delays but—as in the primates—is unable to prevent secondary disease in the majority of animals. A treatment of the donor with ALS and sheep red blood cells delayed the onset of chronic secondary disease significantly more than with ALS alone. A treatment of the donor with thymectomy and ALS led to a 100 day survival of 75% of the recipient mice. These animals did not show clinical signs of chronic secondary disease. Skingrafts of the donor strain placed on the recipients on the 100th day showed a good hair growth 30 days later.
Zusammenfassung
Die Erhöhung der Immunsuppression von Antilymphozytenserum durch zwei weitere Behandlungen des Knochenmarkspenders wurde untersucht. Dabei wurde eine stark H2-unverträgliche Spender-Empfänger-Kombination gewählt, nämlich C57B1/6J auf CBA/J-Empfänger. In dieser Kombination führt ALS zu einer Verzögerung des Auftretens der Sekundärkrankheit aber—in der Mehrzahl der Tiere—genau wie bei Primaten nicht zur vollständigen Unterdrückung der chronischen Sekundärkrankheit. Eine Behandlung des Knochenmarkspenders mit ALS und Hammelerythrozyten verzögerte die chronische Sekundärkrankheit signifikant mehr als mit ALS allein. Eine Behandlung des Knochenmarkspenders mit Thymektomie und ALS führte zu einer Überlebenszeit von 75% der Empfängertiere am 100. Tag post transpl. Diese Tiere zeigten keine klinischen Zeichen der chronischen Sekundärkrankheit. Hauttransplantate vom Spenderstamm, die ihnen am 100. Tag transplantiert worden waren, zeigten einen uneingeschränkten Haarwuchs von der Haarfarbe des Spenderstammes.
