Hämoglobingehalt und Hämoglobinkonzentration in Erythrozyten nach Schichtungszentrifugation

Zusammenfassung

Von Erythrozyten des Erwachsenen- und Nabelschnurblutes wurden nach Zentrifugation in einer schmalen hohen Säule die Zellen der oberen und unteren Schicht isoliert und in ihnen quantitative Daten ermittelt. Die Erythrozyten der unteren Schicht besitzen im Vergleich zu den Zellen der oberen Schicht einen signifikant höheren mittleren Hämoglobingehalt, eine höhere mittlere Hämoglobinkonzentration und ein niedrigeres mittleres Zellvolumen. Die Umlagerung der Erythrozyten während der Zentrifugation ist demnach im wesentlichen eine Funktion der größeren Dichte von Einzelzellen der Gesamtpopulation. Im Hinblick auf die Alterstrennung konnte an Hand der Zahl der Retikulozyten sowohl für Erwachsenen- als auch für Nabelschnurblut eine sichere Anreicherung von jungen Zellen in der oberen Schicht bestätigt werden. Ferner ließ sich für das Blut von jungen Säuglingen bzw. Neugeborenen durch quantitative Bestimmung der Erythrozyten mit adultem Hämoglobin eine Anhäufung von alten Erythrozyten in der untersten Zellschicht nachweisen.

Summary

Erythrocytes of adult and cord blood were centrifuged. In a narrow, high column the cells of the upper and lower layer were isolated and quantitative data were determined within them. The erythrocytes of the lower layer as compared to the cells of the upper layer have a significantly higher mean hemoglobin content, a higher hemoglobin concentration and a lower mean cell volume. Therefore, the rearrangement of erythrocytes during centrifugation essentially is a function of the greater density of individual cells of the total population. With regards to age separation, the number of reticulocytes both in adult and in cord blood confirmed a definite accumulation of young cells in the upper layer. Quantitative determination of erythrocytes in the blood of infants or newborns with adult hemoglobin disclosed an accumulation of old erythrocytes in the lowest cellular layer.
